Switching Nitrofurantoin Polymorphic Outcome in Solvent-Mediated Phase Transformation and Crystallization Using Solvent and Additives

Abstract: We demonstrate that several additives and solvents allow switching the polymorphic outcome of solvent-mediated phase transformation experiments and crystallization of the antibacterial drug nitrofurantoin. Polymorph β is obtained from most of the solvents, whereas the selection of alcohols as solvents or the use of crystallization additives provides the formation of polymorph α. We also demonstrate that this can be linked to reversed apparent relative solubility of nitrofurantoin polymorphs in these solvents o… Show more

“…Intermolecular interactions analysis, thermodynamic analysis, and quantitative calculation as promising and useful tools for polymorph exploration are commonly used to assess the stability and the structure–activity relationship of target crystalline forms, as well as to gain insight into the mechanism of transformation between them [ 36 , 37 , 38 , 39 , 40 ]. Meanwhile, the solid-state transformations (SST) [ 41 , 42 , 43 ] and solution-mediated polymorphic transformations (SMPT) [ 44 , 45 , 46 , 47 ] experiments were usually designed to investigate the transformation mechanism between the crystal forms in recent research.…”

“…While the solubility differential between metastable and stable phases indeed forms the bedrock of the polymorphic transformation process, it is but one piece of the intricate puzzle governing the transformation rate [45] , [46] , [47] . Delving deeper into this phenomenon unveils the pivotal role of additives in orchestrating the transformation process [48] , [49] , [50] , [51] , [52] . These remarkable compounds wield the power to hinder the pre-nucleation aggregation growth of the metastable polymorph, thus acting as gatekeepers to its nucleation.…”
