The objective of this work was to prepare PVA/PAA films containing AC at pH 2.0 and 4.0. The results show that the pH of the film preparations influences the polymer¾drug interaction kinetic order and the degree of swelling. The mechanism of release of AC from the films obtained at pH 4.0 was anomalous, whereas for the films prepared at pH 2.0 the release followed zero-order kinetics.Keywords: hydrogel; acyclovir; drug delivery system.
INTRODUÇÃOHidrogéis compõem um grupo de polímeros constituídos por cadeias macromoleculares arranjadas tridimensionalmente, dispondo da característica de se intumescer ou contrair quando exposto a meios com diferentes valores de pH, temperatura, força iônica e campo elétrico. [1][2][3] No entanto, a configuração da estrutura tridimensional dos polímeros empregados na produção de hidrogéis depende dos valores de pH e força iônica da solução no qual são preparados ou imersos, sendo atribuída à conversão da energia interna as alterações no volume destes materiais. 4 Esta característica tem suscitado o interesse da indústria farmacêutica quanto ao emprego de polímeros que formam hidrogéis na fabricação de sistemas de liberação de fármacos, visto que estes materiais não se dissolvem em meio aquoso quando expostos à temperatura e ao pH fisiológico, estando a taxa de liberação do fármaco a partir da matriz polimérica regulada pela densidade das ligações intermoleculares e pelo grau de intumescimento. [5][6][7][8][9] No entanto, aspectos relativos à biocompatibilidade e propriedades mecânicas deste grupo de polímeros também devem ser considerados quando se almeja seu uso na elaboração de sistemas farmacêuticos.
5O poli (vinil álcool) (PVA) é um polímero não tóxico e biocompatível, que apresenta excelente propriedade de formação de filme e disponibilidade de processabilidade, bem como resistência mecânica, térmica e química. Estas propriedades do PVA têm fomentado sua aplicação nas áreas médica e farmacêutica.10,11 Basak et al. elaboraram filmes contendo PVA reticulado com anidrido maleíco, tendo observado que o aumento no pH da solução na qual os filmes foram imersos ocasionou um aumento no grau de intumescimento e no tempo de liberação de dois fármacos modelos empregados no estudo.
