Sweet switching: Development of photoswitchable glycoconjugates ‐ Mini‐Review

Hemipiperazines are a recently discovered class of peptide-derived molecular photoswitches with high biocompatibility and therapeutic potential. Here, for the first time we describe photochromism of heterocyclic hemipiperazines. They demonstrate long thermal lifetimes, and enlarged band separation between photoisomers. Efficient photoisomerization occurs under aqueous conditions, although with a need for organic co-solvent. Bidirectional switching with visible light is observed for an extended aromatic system.

Thanks to supramolecular interactions of macrocyclic compounds with their guest ions or molecules, macrocycles have found wide applications in molecular and chiral recognition, separation, transportation, molecular machines, and so on. Photoswitchable macrocycles are especially appealing and attracting more and more interest because the embedded molecular photoswitch enables dynamic control of molecular shape, conformation, and different properties by light, a non‐invasive, remote and highly tunable stimulus. With available photochromic compounds, various photoswitchable macrocycles have been developed. In this review paper, we describe the recently reported photoswitchable macrocyclic compounds, with a focus on the control of the macrocyclic structure, dynamic, photo or thermal isomerization, as well as their applications on encapsulation and release of ionic or aromatic species, on the reversible modulation of chemical, biological, electronic, fluorescent and chiroptical properties by light.

Molecular photoswitches reversibly convert the energy of light into a variety of structural changes at the molecular level. They can be integrated into smart materials with broad range of applicability. While most photochromic systems require UV light for activation, application of photoswitches operational entirely within the visible or NIR range of light brings considerable benefits, such as biocompatibility, better light penetration through the material, propelling with sunlight, or a possibility to design dual systems that are mutually activated with UV or visible light frequencies for two distinct functions.
