Svetamycins A–G, Unusual Piperazic Acid-Containing Peptides from Streptomyces sp.

Abstract: Seven new halogenated peptides termed svetamycins A-G (1-7) have been isolated from laboratory cultures of a Streptomyces sp. Svetamycins A-D, F, and G are cyclic depsipeptides, whereas svetamycin E is a linear analogue of svetamycin C. Their structures were determined using extensive spectroscopic analysis, and their stereochemical configuration was established by a combination of NMR data, quantum mechanical calculations, and chemical derivatizations. Svetamycins are characterized by the presence of a hydrox… Show more

“…The plates were incubated for 48 h at 37°C, shaking at 180 rpm. Cell viability was determined using the BacTiter-Glo TM assay (Promega, Mannheim, Germany) and a LUMIstar OPTIMA microplate luminometer (BMG Labtech, Ortenberg, Germany) for the readout (Dardic et al, 2017).…”

“…Other PA-containing cyclic hexapeptides contain only three PA moieties, as, for example, the svetamycins (Dardic et al 2017) or NW-G10 and NW-G11 (Wei et al 2012), two PA moieties, as, for example, depsidomycin (Isshiki et al 1990), piperastatin A þ B Murakami, Harada, Yamazaki et al 1996) or pipalamcin (Uchihata et al 2002), or only one PA moiety as in the case for marformycins (Liu et al 2015).…”

“…Piperazic acid (Piz; PubChem CID: 2762538), a cyclic hydrazine and a non‐proteinogenic amino acid, has been found in over 140 molecules of natural products, many of which exhibit promising biological activities (Ciufolini and Xi, 1998; Morgan et al ., 2019): antrimycins (Shimada et al ., 1981) and svetamycins (Dardic et al ., 2017) as tuberculostatic agents, kutznerides (Broberg et al ., 2006) as antimicrobial agents and luzopeptins as anti‐human immunodeficiency viral agents (Ohkuma et al ., 1980). Moreover, Piz has been shown to be able to induce β‐turns in peptides (Ciufolini and Xi, 1998).…”
