Susceptibility to Neutralization by Broadly Neutralizing Antibodies Generally Correlates with Infected Cell Binding for a Panel of Clade B HIV Reactivated from Latent Reservoirs

Abstract: Efforts to cure human immunodeficiency virus (HIV) infection are obstructed by reservoirs of latently infected CD4 T cells that can reestablish viremia. HIV-specific broadly neutralizing antibodies (bNAbs), defined by unusually wide neutralization breadths against globally diverse viruses, may contribute to the elimination of these reservoirs by binding to reactivated cells, thus targeting them for immune clearance. However, the relationship between neutralization of reservoir isolates and binding to correspon… Show more

“…Since ADCC generally correlates with neutralization (1214), we tested PGT145 for the ability to neutralize the same viruses that were susceptible to ADCC. As expected, PGT145 potently neutralized viruses expressing HIV-1 Env proteins, including HIV-1 NL4-3 , HIV-1 JR-FL , and SHIV AD8-EO (Fig.…”

“…Although instances of neutralization in the absence of ADCC have been observed, most antibodies that mediate ADCC against HIV-infected cells also neutralize viral infectivity (1214). Thus, the ability of PGT145 to direct NK cell killing of SIV-infected cells without blocking SIV infectivity is unusual.…”

“…Besides neutralization per se, Fc-mediated functions play an essential role in Ab protection against HIV infection [133]. There is significant overlap between the capacities of Abs to mediate ADCC against HIV-infected cells and to neutralize viral infection [165,166,167,168]. However, Fc-mediated functions may be partially redundant for very potent bnAbs [169], and ADCC can be uncoupled from neutralization for Abs with low-affinity binding to Env [170].…”
