Susceptibility of BAFF-var allele carriers to severe SLE with occurrence of lupus nephritis

Abstract: Background: Dysregulation of the B-cell activating factor (BAFF) system is involved in the pathogenesis of systemic lupus erythematosus (SLE). Increased serum concentrations of BAFF are related to lupus nephritis and disease activity among SLE patients. Recently, a variant of the BAFF-encoding gene, BAFF-var, was identified to be associated with autoimmune diseases, in particular SLE, and to promote the production of soluble BAFF. The present study aimed to assess the prevalence of BAFF-var in a cohort of 195 … Show more

“…There is limited evidence of genetic contribution to specific organ involvement in SLE. 1 , 2 We identify a large deletion in intron 7 of Van Gogh Like 1 ( VANGL1 ), which associates with nephritis in SLE patients. The same deletion occurs at increased frequency in an indigenous population (Tiwi Islanders) with 10-fold higher rates of kidney disease compared with non-indigenous populations.…”

“…26,29 The utilization of immunosuppressive and systemic steroid agents may potentially influence the comorbidities in patients with SLE. [48][49][50] Incidence of cardiovascular was significantly higher in patient with SLE. This study identified atherosclerosis as the primary comorbidity, followed by hypertension, hypercholesterolemia, obesity, and hyperglycemia.…”

“…At the same time, no relationship was found between the BAFF-var allele and disease manifestations in another study. [ 42 , 43 ] Several clinical studies have reported that s-BAFF levels are higher in patients with renal involvement. [ 11 , 15 , 23 , 27 , 44 ]…”
