Survival Tests for Leptospira spp.

Abstract: Measuring viability is an important and necessary assessment in studying microorganisms. Several methods can be applied to Leptospira spp, each with advantages and inconveniencies. Here, we describe the traditional colony-forming unit method, together with two other methods based respectively on the reducing capacity of live cells (Alamar Blue Assay) and differential staining of live and dead cells (LIVE/DEAD BacLight). The Alamar Blue Assay uses the blue reagent resazurin, which can be reduced into the pin… Show more

“…Survival of leptospires upon treatments with antimicrobial peptides was assessed by Alamar Blue viability assay ( 28 ). After 2 h incubation with LL-37 or Bmap28 in PBS, 2.5 × 10 6 leptospires in 100 μL in 96-well plates were mixed with 80 μL of EMJH and 20 μL of 10× Alamar Blue dye.…”

“…Rezasurin (Alamar Blue Assay, ThermoFisher Scientific) was added and cells were further incubated for 24h. Viability is assessed by the reduction of blue resazurin into pink resorufin [ 55 ]. Plating experiments were performed by diluting treated and non-treated cells in EMJH in the absence of H 2 O 2 and plating the samples on EMJH agar medium [ 55 ].…”

“…For the colorimetric assay, 100 µl of leptospires were incubated with resazurin (Alamar Blue reagent, ThermoScientific) for 48h. Leptospires viability was assessed by their capacity to reduce blue resazurin into pink resorufin as described previously [54].…”
