Survival of Penicillium spp. conidia during deep-frying and baking steps of frozen chicken nuggets processing

Abstract: This study aimed at determining whether Penicillium spp. strains could survive through the heat treatment applied during the processing of frozen chicken nuggets. Firstly, it was found that the conidia of Penicillium were not able to survive the heat shock in phosphate buffer at pH 7.2 in thermal death tubes (TDT) at 80 °C/30 min. Subsequently, each Penicillium strain was inoculated in frozen chicken nuggets, which were subjected to the following treatments: i) only deep frying (frying oil at 195-200 °C), ii) … Show more

“…Conidial suspensions of each strain were individually prepared and had their concentration standardized at 10 6 conidia/mL according to Wigmann, et al . 58 . A. carbonarius and A. niger strains were inoculated in MEA- Malt Extract Agar medium (Acumedia) (Malt Extract: 20.0 g, Peptone: 1.0 g, Glucose: 30.0 g, Agar: 20 g, Distilled Water: 1 L) and incubated at 25 °C for 7 days.…”

“…These strains were previously selected in grape-based culture medium . Conidia suspensions of each species were prepared individually . Once suspensions were obtained, the final concentration of the inoculum was standardized at 10 7 conidia/mL.…”

“…This strain was isolated from soil in Uganda as reported by Singh and Cotty (2019). The strain was prepared according to Wigmann et al (2016) by inoculating it in Malt Extract Agar (MEA; Acumedia, Lansing, Michigan, United States; Malt Extract: 20.0 g; Peptone: 1.0 g; Glucose: 30.0 g; Agar: 20 g; and Distilled Water: 1 L) and incubating it aerobically at 25°C in a BOD incubator for 5 days, at static condition in the dark. Conidia were collected by scraping the mycelium from each plate with sterile distilled water containing 0.1% Tween 80 (Labsynth, Diadema, Brazil).…”
