Surface modification of polystyrene biochip for biotin‐labelled protein/streptavidin or neutravidin coupling used in fluorescence assay

Abstract: A polystyrene biosensing chip to be used as a medical diagnostic tool has been developed. Its surface functionalization is optimized to bind the bioanalytes with high efficiency. Coatings of allyldextran monolayers were carried out on polystyrene chips activated with g-irradiation. The effect of the concentration of allyldextran solution on the coating efficiency was studied by surface-sensitive x-ray photoelectron spectroscopy (XPS) and contact-angle measurements. In a second step, sodium periodate chemistry … Show more

“…Pure streptavidin should have a nitrogen content of 12% at 398 eV. [26] However, it has to be considered that streptavidin is expected to be only immobilized inside the pores while the surface of the honeycomb-structured porous film consisted mainly of polystyrene. The nitrogen content measured should therefore be below the nitrogen content of the protein.…”

“…Rapid advancements in biosensor technology over the past several decades has led to widespread implementation across a variety of application fields, ranging from biodefense (Hindson et al, 2005;Ivnitski et al, 2003;Lim et al, 2005;Peruski and Peruski, 2003), to medical diagnostics (Andreotti et al, 2003;Gao et al, 2004), to environmental monitoring (Baeumner, 2003;Farre and Barcelo, 2003). The diversity of applications and sample matrices results in a nonuniform set of operational requirements.…”
