Surface Modification of Multiple Bioactive Peptides to Improve Endothelialization of Vascular Grafts

Abstract: Endothelialization is an effective approach to prevent thrombus formation and enhance vascular graft survival. Surface modification of biomolecules has been proved to be effective in regulating endothelial cell behaviors. In this study, several peptides including YIGSR, RGD, and REDV sequences are covalently immobilized on the surface of electrospun silk fibroin scaffolds and the effects of combined application of these peptides on cell behaviors are studied. The results show that, compared with the scaffolds … Show more

“…Fibers with smaller diameters offer a higher surface area, which can support a more homogenous cell distribution and spreading (27). However, some studies have demonstrated that small variations in diameter of fibers, such as those observed in the present study, do not modify the interaction of cells with the scaffolds (9,28). Therefore, it is believed that the differences in cell morphology obtained for the two groups of scaffolds have been caused mainly by their different rates of TMC and LA.…”

“…The micropore structure in the wall of ASDBVs is considered to be the premise of endothelialization , which is recognized as one of the most effective methods of long‐term anti‐thrombosis of vascular substitutes . At present, the simple and rapid method to achieve endothelialization of ASDBVs has been the focus of many researchers .…”

“…The Arg-Gly-Asp (RGD) peptide, present in fibronectin and responsible for strong cell adhesion, is very efficient in promoting EC adhesion and their migration in vascular grafts [ 75 ]. Moreover, the combination of RGD with peptides occurring in the laminin β chain, such as YIGSR, significantly enhances EC proliferation [ 76 ]. GFPGER, a protein sequence occurring in collagen type I, increases endothelialization more efficiently than collagen and laminin and even reduces platelet adhesion in an ex vivo arteriovenous shunt model under antiplatelet therapy [ 73 ].…”
