Surface-enhanced Raman spectroscopy of blood serum based on gold nanoparticles for tumor stages detection and histologic grades classification of oral squamous cell carcinoma

Abstract: BackgroundTumor stages detection and histologic grades classification are essential for the diagnosis and prognosis of oral squamous cell carcinoma (OSCC). In this research, we apply surface-enhanced Raman spectroscopy (SERS) of blood serum to detect the tumor stages and histologic classification of OSCC.MethodsAccording to TNM classification and World Health Organization histologic grading system, the blood serum samples were collected from a total of 135 OSCC patients in the different tumor stages and histol… Show more

“…Xue and Wang have proved that the SERS technique can be applied to analyze and discriminate the OSCC and even the different stages of the tumor. Gold NPs can enhance the intensities of the SERS spectrum enormously when they were added to the serum sample [ 73 ]. A customized gold-NP-reduced graphene-oxide- (AuNPs-rGO-) based bioelectrode has been prepared as an immunosensing platform for electrochemical detection of oral cancer [ 74 ].…”

“…Interestingly, the level of hypoxanthine in SERS spectrum was reduced in CRC samples when compared with healthy control, which is consistent with the result of metabolomic profiling . The thymine signal at 741 cm −1 is also found to be generally stronger in the healthy group . The lower levels of hypoxanthine in the circulating blood imply that the highly proliferative nature of CRC cells necessitates large quantities of these molecules for transmission of adenosine triphosphate (ATP) and synthesis of nucleic acids .…”

“…The strong bands found at 1141, 1263 and 1484 cm −1 are related to lipids in the blood serum or extracellular vesicles . Lipids found in blood include total cholesterol, high‐density lipoprotein (HDL) cholesterol, low‐density lipoprotein (LDL) cholesterol or triglycerides.…”

“…Silver or gold colloids are utilized as SERS substrates more often than nanostructured surfaces, possibly because of the facile synthesis of colloidal NPs and the simplicity of adsorbing biomolecules on metal surface by mixing. Statistical analysis methods such as PCA, PCA-linear discriminate analysis (PCA-LDA), Support Vector Machine (SVM), and SVM in combination with PCA (PCA-SVM) have been employed to develop effective diagnostic algorithms for classification of SERS spectra between normal and cancer patients 164-166 and for classification of patients with different tumor stages 153, 162.…”
