Surface engineered antifouling optomagnetic SPIONs for bimodal targeted imaging of pancreatic cancer cells

Abstract: Targeted imaging contrast agents for early pancreatic ductal adenocarcinoma diagnosis was developed using superparamagnetic iron oxide nanoparticles (SPIONs). For phase transfer of SPIONs, the hydrophobic SPIONs are first treated with tetrafluoroborate and then capped by bovine serum albumin (BSA) via ligand exchange. It was experimentally found that nitrosyl tetrafluoroborate pretreatment and proper structures of molecules are essential to the effective surface functionalization of SPIONs. Nonspecific binding… Show more

“…The cytotoxicity test of NPs on PANC‐1 cells was performed by different methods . First, the LC 50 results show that after 48 hours of exposure the toxicity for both SiO 2 ‐Fe 3 O 4 NPs and Fe 3 O 4 NPs (around 150 μg/mL) was within a biologically accepted standard deviation for both materials (see Figure ), despite the SiO 2 ‐Fe 3 O 4 NPs show a higher variance in the low concentrations, which is comparable with previous literature .…”

“…To prepare these bioprobes, the obtained core-shell NRs were surface-modifi ed using 1,2-ethanedithiol and bovine serum albumin (BSA) based on previously reported methods. [ 61 ] Curve III in Figure 6 [ 59,60 ] After being transferred, the obtained colloidal aqueous solution was stable with very few precipitation for 3 days ( Figure S1, Supporting Information).…”

“…BSA and its derivatives have numerous biochemical applications that include blocking agents in enzyme-linked immunosorbent assay [30], stabilizers in enzymes processing [31], ligands in surface engineering [24,32] and nanocarriers [33e35]. In this study, BSA has two roles, namely the ligands for surface engineering of hydrophobic QDs and nanocarriers for linking Gd DTPA and antibodies.…”

“…In this study, BSA has two roles, namely the ligands for surface engineering of hydrophobic QDs and nanocarriers for linking Gd DTPA and antibodies. Practically, BSA, as a multivalent ligand with abundant free carboxyl, amino and disulfide bonds, has been successfully used in our previous work for surface engineering of nanopaticles [24,32]. In this study, for multipurpose applications, BSA is employed with MRI moiety beforehand, and utilized for water-solubilization of hydrophobic QDs.…”
