Surface-Attached PDMAA−GRGDSP Hybrid Polymer Monolayers that Promote the Adhesion of Living Cells

Abstract: Peptide-polymer hybrid molecules are being introduced, where one part of the molecule (i.e., the peptide) promotes the adhesion of living cells, whereas the other part of the molecule (i.e., the synthetic polymer) is known to prevent cell adhesion. The hybrid copolymer, poly(dimethylacrylamide) (PDMAA)-glycine-arginine-glycine-aspartic acid-serine-proline (GRGDSP) was synthesized by first preparing an initiator-modified peptide and in a second step growing the PDMAA block directly off the peptide through atom … Show more

“…Proteins present in the serum first adsorb onto the surface presumably via hydrophobic‐hydrophobic interactions and form a monolayer on the polymer network. The adsorbed proteins then mediate interactions (via α,β‐integrins mediated cell adhesion) with adhesion sites on the surface of the cells …”

“…The resulting films allow a spatial adhesion control of human skin fibroblast cells and thus might constitute a platform to design live-cell biochips. [67] Moreover, the synthetic polymer block of bioconjugates can be used to integrate selective polymer functions e.g., to generate switchable surfaces modifications. [68] Börner and coworkers, synthesized a bioconjugate by a ''grafting from'' strategy using a peptide-based chain transfer agent (CTA-GGRGDS) and RAFT polymerization.…”

“…[1,2] Mostly, this has been achieved through the natural adhesion of molecules such as oligo-or poly-saccharide ligands [3] to, for example, pili [4] and selectins, [5,6] or by direct interaction of ligands, such as RGD with receptors involved in the control of cellular outgrowth and migration. [7][8][9][10][11][12][13][14][15] Oligosaccharides, peptides, long hydrophobic chains, and even proteins [16] have been applied to various surfaces, such as metals, glass, and polymeric beads, and have been found to mediate cell binding.…”
