“…In this pathway of apoptosis, TNF-α or other death receptor ligands can activate death receptors on the proximal tubule cell surface leading to caspase 8 cleavage and activation followed by the cleavage and activation of downstream effector caspases such as caspase 3, leading to apoptosis (52). Suramin has been shown to inhibit TNF-α from binding with the death receptor (33). Interestingly our results show that pre-treatment with suramin prior to cisplatin treatment significantly decreases both caspase 8 and caspase 3 cleavage, suggesting that suramin is preventing cisplatin-induced death receptor-mediated apoptosis in the kidney, while suramin pretreatment also decreased the renal expression of TNF-α in mice treated with cisplatin.…”
