Supramolecular Assembly of Peptide Amphiphiles

Abstract: ConspectusPeptide amphiphiles (PAs) are small molecules that contain hydrophobic components covalently conjugated to peptides. In this Account, we describe recent advances involving PAs that consist of a short peptide sequence linked to an aliphatic tail. The peptide sequence can be designed to form β-sheets among the amino acids near the alkyl tail, while the residues farthest from the tail are charged to promote solubility and in some cases contain a bioactive sequence. In water, β-sheet formation and hydrop… Show more

“…Peptide amphiphiles (PAs) are amphiphilic molecules that contain bioactive peptides. PAs can be synthesized by linking amino acids into hydrophilic and hydrophobic domains, or by conjugating oligopeptide to hydrophobic hydrocarbon chain (lipopeptide) . Due to their amphiphilic properties, PAs self‐assemble into nanostructures under certain conditions.…”

“…Additionally, the assembly and disassembly of the tertiary structures to form the quaternary structure occur reversibly in response to environmental changes that induce dynamic functions. This multistep bottom‐up approach and reversible assembly/disassembly are crucial concepts for fabrication of the sophisticated hierarchical structures . To realize reversible multistep assembly, the design of the primary structure is important.…”

“…A well established technique that is, lipidation of peptide produced a binary system called peptide amphiphile, an approach to mimic functioning of cellular protein and its permeability for membrane of microorganisms which is formed by long chain fatty acids . Lipid‐peptide conjugation performed a wide diversity in the sequence of modulation of peptide self assembly …”

“…It is obvious that, upon suspending the sPA in a polar solvent, the lipid chains are attracted towards each other and away from polar solvent while the polar part (head) stabilizes by moving towards polar solvent and the aromatic section helps into self organisation followed by stabilization of sPA nanostructres through π–π interaction . Therefore, the shape of this self‐assembly primarily dependent on the nature of the hydrophilic head and used aromatic dipeptide(s) …”
