Suppressor of RNA silencing encoded by Beet yellows virus

Abstract: Using an Agrobacterium-mediated transient assay, we screened the 15.5-kb genome of the Beet yellows virus for proteins with RNA silencing suppressor activity. Among eight proteins tested, only a 21-kDa protein (p21) was able to suppress double-stranded (ds) RNA-induced silencing of the green fluorescent protein (GFP) mRNA. Restoration of GFP expression by p21 under these conditions had no apparent effect on accumulation of the small interfering RNAs. In addition, p21 elevated the transient expression level of … Show more

“…For example, the potyviral HcPro most likely inhibits unwinding of the siRNA duplex and RISC assembly (10), P25 of Potato virus X suppresses the production or accumulation of mobile silencing signals (15), and P19 of Tomato bushy stunt virus (TBSV) and P21 of Beet yellow virus inhibit RNA silencing by physically interacting with siRNAs and preventing their processing or incorporation into the RISC (9,10,(16)(17)(18)(19)(20). However, among numerous known viral RNA-silencing suppressors, only two, the 2b protein of Cucumber mosaic virus (CMV) and poleroviral (P0), have been shown to interact directly with a protein component of the host-silencing pathway.…”

“…activity of p20 was first suggested in the study that identified the p20 homolog of Beet yellows closterovirus as a silencing suppressor (10,35). p20 suppression shares features with silencing suppression by CMV 2b because both are potent suppressors of intercellular silencing but incomplete in suppressing intracellular silencing (21).…”

“…To determine whether the inhibition of recombinant TYMV replication would be relieved by viral RNA silencing suppressors, we introduced the TY-GUS construct into N. benthamiana along with potyviral HC-Pro from Tobacco etch virus (18), closteroviral p21 from Beet yellows virus (19), or tombusviral p19 from Tomato bushy stunt virus (20). Using this strategy, the replication of the recombinant TYMV recovered to the wildtype level when one of the viral RNAi suppressors was co-expressed ( Fig.…”
