Suppression of SCIN inhibits human prostate cancer cell proliferation and induces G0/G1 phase arrest

Abstract: DNA polymerase kappa (POLkappa) is a low fidelity translesional DNA polymerase implicated in spontaneous and DNA damage-induced mutagenesis. We have previously shown that POLkappa was frequently overexpressed in human lung cancer tissues as compared with their matched non-tumorous tissue counterpart. In the present study, we found a close correlation between elevated POLkappa expression and p53 inactivation in lung cancer tissues. To investigate whether POLK expression might be regulated by p53, we have determ… Show more

“…Previous studies suggest that Mith inhibits growth of human cancers by decreasing Sp1 protein232425, but how Sp1 protein is decreased by Mith is not clear. We checked Sp1 mRNA levels in cervical cancer cell lines and saw no significant difference in cells with or without Mith treatment (Fig.…”

“…Previous studies have suggested that Mith inhibits the growth of various human cancers by decreasing Sp1 protein level232425, but it is still not clear how Sp1 protein level is decreased by Mith. We first checked the mRNA expression levels of Sp1 in different cervical cancer cell lines.…”

“…After 1, 2, 3, 4, and 5 days of incubation, viable cells were analyzed using MTT plus acidic isopropanol (10% SDS, 5% isopropanol, and 0.01 M HCl), as described in a previous report. 19 The absorbance of each well was recorded at the wavelength of 595 nm using a microplate reader. Experiments were performed in triplicate.…”

“…79, 80, 83–85 For example, higher stage gliomas have been found to over-express DNA damage tolerance components. 86 Over-expression of hpol Îș was observed in 23 out of 40 glioma patients (57.5%) and hpol Îč in 11 of 40 (27.5%), while there was no observable change in hpol η expression between glioma patients and normal tissue.…”
