Suppression of Oxidative Stress as a Mechanism of Reduction of Hypercholesterolemic Atherosclerosis by Cyclooxygenase Inhibitors

Abstract: Hypercholesterolemia increases the formation of arachidonic acid from membrane phospholipids. Reactive oxgyen species (ROS) are generated during the synthesis of prostaglandins (PGs) from arachidonic acid. ROS have been implicated in the development of hypercholesterolemic atherosclerosis.

“…Aortic-CL levels in group II were similar to those in group I. Aortic-CL in rabbits decreases with 0.25% cholesterol 25,35 and 0.5% cholesterol diets, 5,22 and increases with a 1% cholesterol diet. 4 No change in aortic-CL in the present study could be due to the 1% cholesterol diet for 1 week in the fourth week of the protocol.…”

“…Arterial blood collected in ethylenediamine tetraacetic acid-containing tubes was used to measure WBC counts using a Technicon H6000 System (Technicon Instruments Corp, Terrytown, NY), and the WBC-CL was measured using a previously described method. 1,22 Briefly, 0.05 mL of blood was added to a tube containing Hank's balanced salt solution (HBSS), and luminol at a final concentration of 10 -4 mol/L. Samples were placed in a luminometer (AutoLumat LB953) at 37 C and phagocytosis was initiated by adding 0.1 mL (10 mg/mL stock solution) of opsonized zymosan, prepared as described previously.…”

“…[1][2][3][4][5] Antioxidants and suppressants of sources of ROS are effective in the suppression of the development of atherosclerosis. [1][2][3][4]6,7 Flax lignan complex contains secoisolariciresinol diglucoside (SDG; 34% to 38%), cinnamic acid glucoside (15% to 21%), and hydroxymethylglutaric acid (9.6% to 11.0%). 8 SDG 9,10 and cinnamic acid 11 are antioxidants.…”

“…Hypercholesterolemia induces oxidative stress, since it increases the formation of reactive oxygen species from membrane phospholipids during prostaglandin synthesis. Thus antioxidants and hypolipidemic agents suppress the development of hypercholesterolemic atherosclerosis and induce regression of atherosclerosis (Paul and Kailash, 2003). Therefore suppression of atherosclerosis is associated with decrease in oxidative stress and serum lipids (Kabiri et al, 2011).…”
