Suppression of microRNA-18a expression inhibits invasion and promotes apoptosis of human trophoblast cells by targeting the estrogen receptor α gene

Abstract: The purpose of the present study was to gain further understanding of the function of microRNA-18a (miR-18a) expression in the JEG-3 human trophoblast cell line. JEG-3 cells were transfected with pre-miR-18a mimics or miR-18a inhibitors. The effects of miR-18a on trophoblast cell invasion and apoptosis, and on the expression of estrogen receptor α (ESRα) were analyzed using a Transwell invasion assay, flow cytometry, reverse transcription-polymerase chain reaction, western blot analysis and a luciferase assay.… Show more

“…Thbs1 (thrombospondin 1) is an effective angiogenesis inhibitor 28 whose expression is inhibited by Myc protein 23 and Myc-induced miR-17/92 cluster miRNAs, 29 in particular, miR-18a and miR-19b-1. 30 32 To assess whether Thbs1 expression was modulated in E9.5 Sema3f −/− yolk sacs compared with their wild-type counterpart, we first analyzed Thbs1 gene transcription by real-time quantitative polymerase chain reaction (Figure IVB in the online-only Data Supplement ). Compared with wild-type yolk sacs, Thbs1 mRNA levels increased by 1.5-fold in Sema3f −/− yolk sacs.…”

“…Several reports have shown that miR-18a-5p is related to cell death. Suppression of miR-18a can increase apoptosis ( Fujiya et al ., 2014 ; Zhu et al ., 2015 ). In addition, miR-18a-5p may promote carcinogenesis by directly targeting IRF2 in lung cancer ( Liang et al ., 2017 ) and STK4 in prostate cancer ( Hsu et al ., 2014 ).…”

“…The miR-29 family (miR-29a/b/c) promoted apoptosis by targeting myeloid cell leukemia-1 (MCL1), an apoptosis regulator and a member of the BLC2 family , Gu et al 2016. Overexpression of miR-18a increased apoptosis by inducing the expression of estrogen receptor alpha (ESR1) (Zhu et al 2015), while miR-128a induced apoptosis via the mitochondrial pathway by downregulating BAX (Ding et al 2016) and miR-30a-3p by inhibiting IGF1 (Niu et al 2018). On the other hand, miR-101 targeted endoplasmic reticulum protein 44 (ERP44) to suppress ER-stress-induced apoptosis (Zou et al 2014).…”
