Suppression of arthritic bone destruction by adenovirus‐mediated dominant‐negative Ras gene transfer to synoviocytes and osteoclasts

Abstract: Conclusion. Ras-mediated signaling pathways are involved in the activation of RA SFCs and the destruction of bone in arthritic joints, suggesting that inhibition of Ras signaling can be a novel approach for RA treatment that targets both synovial cell activation and bone destruction in the RA joint.Rheumatoid arthritis (RA) is a chronic, systemic inflammatory disease of unknown etiology that is characterized by invasive synovial hyperplasia, leading to progressive joint destruction. Rheumatoid synovial cells a… Show more

“…This suggests that SD-282 healed bones in the late stage of CIA disease by reducing bone resorption and by forming new bone. We hy- pothesize that the effect of SD-282 on bone may be due to inhibition of osteoclast differentiation in the absence of IL-6 (Li et al, 2002(Li et al, , 2003Yamamoto et al, 2003) and/or osteoblastogenesis. We are in the process of testing the latter hypothesis.…”

“…Synovial cells were obtained as previously described (13,23,24). In brief, with enzymatic digestion, human synovial cells were isolated from synovial tissues of the knee joints of ten rheumatoid arthritis patients (37-75 years of age; mean, 60.3 years of age) at the time of total knee arthroplasty operations.…”

“…Such inhibitors also reduce severity of collagen-induced arthritis (an animal model of RA) by suppressing Ras and IKK-mediated activation of NF-B (53). Previously observed suppression of bone destruction by dominant negative Ras may be partly due to inhibition of proteases such as ADAMTS-4 and MMPs (54,55).…”
