Background and Aim: Brucellosis is considered as an important zoonotic disease caused by various strains of Brucella in numerous host species. Although brucellosis has been reported in almost animal species, the relevance of brucellosis infection and diagnostic technique in Asian elephant (Elephas maximus) has been limited. The present serological investigation aimed to investigate the antibody response to Brucella abortus in captive Asian elephants in North Thailand. Moreover, further serological survey was also conducted to detect the antibody response to Brucella canis in stray dogs cohabiting the same area as the elephant herd. Materials and Methods: Serum samples were collected from 40 captive Asian elephants and submitted for serological analysis based on B. abortus antigen using Rose Bengal plate test (RBPT) in combination with ethylenediaminetetraacetic acid-tube agglutination test (EDTA-TAT) as a supplementary test and by commercial indirect enzyme-linked immunosorbent assay (iELISA). In addition, serum samples were also obtained from 16 stray dogs that live nearby the elephant-raising area and were tested using commercial Dot-ELISA based on B. canis antigen. Results: Serological analysis in captive Asian elephants showed 100% seronegative (40/40) from all serological tests response to B. abortus. For stray dogs, 12.5% (2/16) had a low positive reaction response to B. canis. Conclusion: The serological survey for brucellosis in Asian elephant was adapted and applied using RBPT, EDTA-TAT, and iELISA in the present study. For future evaluation, we recommended the use of a combination of serological tests with validation together with comparing by direct detection such as bacterial isolation to provide an appropriate brucellosis surveillance program in Asian elephants. In addition, the surveillance of stray dogs or multispecies habitation should be kept into considerations.

Relationship between insulin-like growth factor-1 and semen quality in Asian elephants (elephas maximus)

Evidence has been increasingly suggested that insulin-like growth factor-1 (IGF-1) is related to steroidogenesis and spermatogenesis. This may be conducive to find the causes of poor sperm quality in Asian elephants (Elephas maximus). The present study aimed to investigate whether correlations among serum IGF-1 concentration, serum testosterone level and semen variables exist in elephants. A total of 17 ejaculates (1–3 ejaculates/bull) were collected by performing transrectal massage. Before each ejaculate, blood samples were obtained (n = 17). Subsequently, semen characteristics of each ejaculate were evaluated. Assessments of precision (calculations of the intra- and inter-assay coefficient of variation, CV) and accuracy (tests of linearity and spike-recovery) were performed for ELISA validation. An increase of serum IGF-1 concentration was found to correlate with the percentages of spermatozoa with intact acrosome (r = 0.53, P < 0.05) and normal head morphology (r = 0.48, P < 0.05). The serum IGF-1 concentration was positively correlated with serum testosterone level (r = 0.73, P = 0.004). The results of validation demonstrated the CV was 1.6–6.4% for intra-assay variability and 4.7–6.9% for inter-assay variability. Linearity under serial dilutions of a known serum concentration was confirmed (R2 = 0.99) with an acceptable recovery rate of each dilution (mean 114.7 ± 19%, ranging from 100 to 143%). Additionally, the mean percentage of recovery of spiked IGF-1 was 107.2 ± 21% (ranging from 80.8 to 136.9%). In summary, this commercial ELISA kit can be used to determine serum IGF-1 concentration in Asian elephants. Moreover, our findings suggest the increased IGF-1 may be relate to good sperm quality and that the relationship between serum IGF-1 and testosterone concentration indicates a crucial role in the fertility of male elephants. Further works are interesting to investigate the exact mechanism by which IGF-1 affects semen quality in elephants.

Context The removal or supplementation of ejaculates with seminal plasma (SP) can affect cryotolerance and post-thaw survival of spermatozoa in many species. In the Asian elephant (Elephas maximus), elucidation of the SP proteome and investigation of how it affects spermatozoa may enable improvement of cryopreservation protocols. Aims Herein, we characterise the Asian elephant SP proteome and investigate the impacts of SP on sperm cryotolerance in the presence of conspecific or heterospecific SP. Methods Proteomic analysis of Asian elephant SP was performed using mass spectrometry on nine samples from three individuals. In a separate study, SP was removed from six ejaculates and spermatozoa were resuspended in Tris extender supplemented with: no seminal plasma (NOSP), conspecific SP from ejaculates exhibiting ‘good’ (GSP, >60%) or mixed sperm total motility (MSP), or horse SP (HSP). Samples underwent cryopreservation, and sperm parameters were compared prior to cryopreservation and after thawing (0 and 2 h). Key results Mass spectrometry identified 155 proteins from an array of families. Significant differences were observed in post-thaw sperm quality between SP treatments: high concentrations of MSP (25%, v/v) displayed greater average path and straight-line velocity immediately after thawing (P < 0.05) and greater sperm motility index and beat cross frequency than NOSP after 2 h post-thaw incubation (P < 0.05). The addition of HSP improved sperm kinematic parameters compared to NOSP and GSP treatments (P < 0.05). Conclusions and implications These preliminary findings suggest the potential of SP to enhance the cryosurvival of Asian elephant spermatozoa, with HSP showing particularly promising results compared to conspecific SP (GSP). Further research into the specific effects of Asian elephant SP proteins is warranted.
