BackgroundDouchi (fermented Glycine max Merr.) is produced from fermented soybeans, which is widely used in traditional herbal medicine. In this study, we investigated whether Douchi attenuates protein kinase C (PKC) and interleukin (IL)-4 response and cutaneous inflammation in Atopic dermatitis (AD)-like NC/Nga mice.MethodsTo induce AD-like skin lesions, D. farinae antigen was applied to the dorsal skin of 3-week-old NC/Nga mice. After inducing AD, Douchi extract was administered 20 mg/kg daily for 3 weeks to the Douchi-treated mice group. We identified the changes of skin barrier and Th2 differentiation through PKC and IL-4 by immunohistochemistry.ResultsDouchi treatment of NC/Nga mice significantly reduced clinical scores (p < 0.01) and histological features. The levels of PKC and IL-4 were significantly reduced in the Douchi-treated group (p < 0.01). The reduction of IL-4 and PKC led to decrease of inflammatory factors such as substance P, inducible nitric oxide synthase (iNOS) and Matrix metallopeptidase 9 (MMP-9) (all p < 0.01). Douchi also down-regulated Th1 markers (IL-12, TNF-α) as well as Th2 markers (IL-4, p-IκB) (p < 0.01).ConclusionDouchi alleviates AD-like skin lesions through suppressing of PKC and IL-4. These results also lead to diminish levels of substance P, iNOS and MMP-9 in skin lesions. Therefore, Douchi may have potential applications for the prevention and treatment of AD.

Atopic dermatitis (AD), which is known as the most common pruritic skin disease, is caused by epidermal barrier dysfunction, allergies, microwave radiation, histamine intolerance, and genetic defects. To investigate the therapeutic effects of fermented soycrud (FSC) on AD pathology, alteration of AD phenotypes induced by phthalic anhydride (PA) treatment was assessed by ear thickness analysis, measurement of immune-related organ weights, ELISA, and histological and pathological analyses of ICR mice after FSC treatment for 2 weeks. Except for water content, the concentrations of most major components were lower in FSC compared to common tofu (CMT). Thymus and lymph node weights were significantly reduced in ICR mice treated with PA+CMT or PA+FSC, whereas spleen and body weights were maintained. Elevation of ear thickness induced by PA treatment was rapidly diminished in the CMT- and FSC-treated groups, although there was no significant difference between the two groups. Furthermore, significant reduction of epidermal thickness was detected in both the PA+CMT- and PA+FSC-treated groups. However, IgE concentration and dermal thickness were reduced only by PA+FSC treatment, whereas PA+CMT treatment maintained levels comparable to PA+vehicle treatment. The number of infiltrated mast cells was higher in the PA+vehicle-treated group compared to the untreated control. Following CMT or FSC treatment, mast cell infiltration was slightly reduced, although the CMT-treated group showed greater cell numbers. These results indicate that FSC may significantly relieve the phenotypes of AD induced by PA treatment and should be considered as a potential candidate for AD therapy.

Antioxidant and Prebiotic Activity of Selected Edible Wild Plant Extracts

Edible wild plants were investigated as potential sources of antioxidants and prebiotics to benefit human health. Antioxidant activity, ascorbic acid and total dietary fibre contents were determined in edible wild plants from Lesotho, Swaziland and South Africa. Pure probiotic strains of Bifidobacterium animalis subsp. animalis (ATCC 25527), Lactobacillus rhamnosus (TUTBFD) and Lactobacillus acidophilus (ATCC 314) were cultured in broth containing edible wild plant extracts to assess their prebiotic activity. Cyperus esculantus had the highest arscobic acid content of 603±64.1 mg/100 g edible dry plant material followed by Rosa rubiginosa (500.8±48.8 mg/100 g). The two plants had IC 50 of 10.7±0.2 µg/mL and 47.8±0.2 µg/mL for DPPH inhibition, respectively. Forty percent (40%) (n=30) of the edible wild plants had significant (p<0.01) total antioxidant activity (IC 50 <60 µg/mL) and high ascorbic acid content (>200 mg/100 g). Nasturtium officinale reported the highest yield for soluble fibre (25%) while Hypoxis hirsute had the highest total dietary fibre content (7.3%). Rorippa nudiuscula enhanced the growth of B. animalis significantly (p=0.001), 8-fold more than inulin. Chenopodium album and Urtica dioica stimulated the growth of L. rhamnosus significantly (p=0.0001) than inulin, respectfully, while Tragopogon porrifolius significantly (p=0.0001) stimulated the growth of L. acidophilus than inulin. It was concluded that the investigated edible wild plants from southern Africa have antioxidant and prebiotic properties that may be beneficial to human health.
