Supplementation ofBacillus amyloliquefaciensAS385 culture broth powder containing 1-deoxynojirimycin in a high-fat diet altered the gene expressions related to lipid metabolism and insulin signaling in mice epididymal white adipose tissue

Abstract: Supplementation of Bacillus amyloliquefaciens AS385 culture broth powder in high-fat diet restored adiposity, glucose tolerance and insulin sensitivity in mice.

“…16−18 However, the working mechanisms behind these effects are still unclear, although some reports have linked the absorption and organ uptake of DNJ and fagomine with their functional properties. 17,19 Considering this, detailed information about the pharmacokinetic profile of aza-sugars is necessary to accurately determine the working mechanism of DNJ, fagomine, and GAL-DNJ.…”

“…Aza-sugars in mulberry leaves (e.g., DNJ and fagomine) have been widely known for their antihyperglycemic activity . More recent studies have started to put emphasis on other beneficial features of DNJ and fagomine, such as antilipid accumulation, antihypertension, and anticancer properties. However, the working mechanisms behind these effects are still unclear, although some reports have linked the absorption and organ uptake of DNJ and fagomine with their functional properties. , Considering this, detailed information about the pharmacokinetic profile of aza-sugars is necessary to accurately determine the working mechanism of DNJ, fagomine, and GAL-DNJ.…”

“…Another report claimed morphine brain accumulation of ~ 0.01% from oral dose of 9 mg/kg 41 . Considering a brain-transportable natural compound, 1-deoxynojirimycin showed an appropriate accumulation of ~ 1.2 pmol/mg in the brain of Sprague–Dawley rats after oral administration at 40 μmol or 6.5 mg/kg 42 , 43 . Conversely, despite the low accumulation or poor uptake ratio of 0.0037% of Tyr-[ 13 C 5 , 15 N]Pro in the mouse brain at 10 mg/kg observed in the present oral administration study, we have previously shown significant improvement in impaired memory in amyloid β-induced AD mice following daily Tyr-Pro intake (100 mg/kg, twice a day) for 16 days 18 suggesting that low Tyr-Pro accumulation, even at 100 mg/kg, would be effective when one expects a preventive effect against the onset of AD.…”

“…1-Deoxynojirimycin (1-DNJ), a polyhydroxylated piperidine alkaloid [ 35 ], is a potent α-glucosidase inhibitor with unique bioactivities, such as anti-inflammatory, antioxidant, and anticancer [ 57 , 58 , 59 ]. 1-DNJ exhibited anti-hyperglycemic activity through regulating the expression of proteins related to glucose transport systems, glycolysis, and gluconeogenesis enzymes [ 60 , 61 , 62 ]. In addition, 1-DNJ also possesses anti-microbial properties.…”
