Superoxide Dismutases

Abstract: Superoxide dismutase was purified from pea (Pisum sativum L., cv. Wando) seeds and corn (Zea mays L., cv. Michigan 500) seedlings. The purified pea enzyme eluting as a single peak from gel exclusion chromatography columns contained the three electrophoreticaUy distinct bands of superoxide dismutase characterizing the crude extract. The purified corn enzyme eluted as the same peak as the pea enzyme, and contained five of the seven active bands found in the crude extract. The similar molecular weights and the cy… Show more

“…The activity of SOD was determined by adding 50 µL of the enzymatic extract to a solution containing 50 µM NBT, 1.3 µM riboflavin, 13 mM methionine, 75 mM EDTA and 50 mM phosphate buffer (pH 7.8) (Giannopolitis & Ries, 1977).…”

“…We determined superoxide dismutase (EC: 1.15.1.1) activity using NBT-illumination [19]. The reaction mixture (3 mL) was composed of 1.5 mL phosphate buffer (pH 7.8), 80 mg methionine (Met), 4 mg nitroblue tetrazolium (NBT), 4 mg Na 2 EDTA, 0.4 mg riboflavin, and 0.1 mL supernatant (i.e., enzyme extract from Z. matrella), and 0.8 mL distilled water.…”

“…Enzyme activity and protein assay 2.3.1. Superoxide dismutase (SOD, EC 1.15.1.1) assay SOD activity was determined according to Giannopolitis and Ries (1977). An assay mix (10 mL) was prepared just before use consisting of 0.5 mM xanthine (prepared fresh at each use), xanthine oxidase diluted with a potassium phosphate buffer (final concentration 0.32 U mL −1 ) prepared afresh at each use and cytochrome c prepared with phosphate buffer and stored at −20°C and on ice during use.…”
