Superior Protection against Malaria and Melanoma Metastases by a C-glycoside Analogue of the Natural Killer T Cell Ligand α-Galactosylceramide

Abstract: α-Galactosylceramide (α-GalCer) is a glycolipid that stimulates natural killer T cells to produce both T helper (Th) 1 and Th2 cytokines. This property enables α-GalCer to ameliorate a wide variety of infectious, neoplastic, and autoimmune diseases; however, its effectiveness against any one disease is limited by the opposing activities of the induced Th1 and Th2 cytokines. Here, we report that a synthetic C-glycoside analogue of α-GalCer, α-C-galactosylceramide (α-C-GalCer), acts as natural killer T cell liga… Show more

“…The cytokine profile of NKT cells is possibly influenced by the quality of the TCR signal, which is dependent on different antigens. An -Galcer analogue with a shortened sphingosine stimulates a higher ratio of IL-4 to IFN- secretion, while a C-glycoside analogue of -Galcer promotes a higher ratio of IFN- to IL-4 (7,8). However, there is no structural basis for predicting how a compound will affect the cytokine profile produced by activated NKT cells.…”

“…Crosstalk between the two cell types involves CD40/CD40L interactions but does not The adjuvant effects of iNKT cells, together with their cytotoxic activities and capacity to secrete diverse cytokines, and the ease by which these cells can be expanded using conserved antigens have placed these cells as important immunomodulators in the tissues and potential targets for immunotherapy. Therapeutic activation of iNKT cells in murine models can prevent tumor growth [182][183][184], ameliorate autoimmune [185,186] and metabolic [187] disease, and protect against microbial infection [188]. Numerical and functional iNKT cell deficiencies have been reported in a number of human diseases [189][190][191][192].…”

“…For example, several synthetic agonist analogs of α-Galcer have distinct effects on iNKT cell function. The OCH [97] and C20:2 [98] analogs have both been shown to elicit a predominantly Th2-like response while α-C-Galcer drives a Th1 response [99]. The reason why these analogs induce different functional responses following iNKT cell stimulation is unclear.…”
