Super-enhancers: critical roles and therapeutic targets in hematologic malignancies

Abstract: Super-enhancers (SEs) in a broad range of human cell types are large clusters of enhancers with aberrant high levels of transcription factor binding, which are central to drive expression of genes in controlling cell identity and stimulating oncogenic transcription. Cancer cells acquire super-enhancers at oncogene and cancerous phenotype relies on these abnormal transcription propelled by SEs. Furthermore, specific inhibitors targeting SEs assembly and activation have offered potential targets for treating var… Show more

“…Super-enhancers are large clusters of enhancers with aberrantly high levels of TFs binding to drive transcription of genes 45 . To find potential TFs that activate expression of DSCAM-AS1, we conducted a motif analysis of the promoter region using ChIPBase 46 and JASPAR 47 .…”

“…Similar long-distance promoter-enhancer interactions exist also in superenhancers (SEs), thus it is not surprising that CDK8 has a role in the regulation of SE-dependent genes. SEs are large clusters of enhancers loaded with particularly high levels of Mediator complex and transcription factors, regulating expression of genes crucial for cell identity and disease, including multiple oncogenes [115][116][117]. Best studied example for CDK8-SE connection was described in acute myeloid leukemia.…”

“…Interestingly, a recent study noted that super-enhancers (SEs) are implicated in specific tumor biology, including EMT 7 . SEs, a large cluster of cis-regulatory DNA elements, can regulate cell-type specific genes by recruiting master transcription factors 8 - 10 . SEs are often identified by histone markers, RNA polymerase 2 (Pol 2) and the transcription cofactor p300 11 - 13 .…”
