Super-assembled core/shell fibrous frameworks with dual growth factors forin situcementum–ligament–bone complex regeneration

Abstract: In this work, a super-assembled framework (SAF) was fabricated to achieve sequential delivery of bFGF and BMP-2 for in situ cementum–ligament–bone complex regeneration.

“…Consequently, as expected, GFs loading could make the iTE-scaffold more biocompatible. The release profiles of iTE-scaffold displayed a sequential release pattern, which was similar to our previous finding [ 20 ]. In brief, bFGF exhibited an initial burst release profile (over 70%) during the first few days.…”

“…0.15 g PLLA and 1 g PLGA were added to 1.25 mL and 5 mL 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP, Macklin, Shanghai, China) so as to make the PLLA core solution and PLGA sheath solution, respectively. After stirring overnight, BMP-2 in phosphate buffered saline (PBS, 0.01 M, HyClone, Logan, USA) (1 μg, 0.125 mL) was added into the core solution by stirring and sonicating, while bFGF in PBS (500 ng, 0.5 mL) was slowly added into the sheath solution in the same way according to our previous study [ 20 ]. In addition, the control group selected PBS as the water phase, which was prepared under similar parameters.…”

“…This material showed new bone formation as well as periodontal ligament and cementum regeneration when implanted in vivo. 485 Others have combined stromal cell derived factor-1 (SDF-1, a chemoattractant) 486 and BMP-2 with hydrogelator Nap-Phe-Phe-Tyr-OH (NapFFY) by simply dissolving and cooling the mixture to induce assembly. 487 Release profiles were steady through one month and results from a maxillary criticalsized periodontal bone defect showed regeneration of periodontal tissue supporting bone.…”

“…In periodontal tissue engineering, recent studies have highlighted the effectiveness of electrospun matrixes to support cell loading, proliferation and differentiation and in vivo periodontal tissue regeneration, in combination with growth factors or cell seeding. Ding et al, created core/shell composite PLLA/PLGA fibrous scaffolds through electrospinning for the dual release of basic fibroblast growth factor (bFGF) and bone morphogenetic protein-2 (BMP-2) [ 47 ]. After eight weeks of implantation in intrabony defects into the alveolar bone crest of Wistar rats, they observed the regeneration of the whole periodontal apparatus (bone, cementum, and periodontal ligament (PDL)).…”
