31 P nuclear magnetic resonance studies of HeLa cells

Abstract: A survey of phosphorus compounds present in HeLa cells and their acid extracts-has been carried out by 31p nuclear magnetic resonance spectroscopy at 40 MHz. The proton decoupled 31p spectrum of the neutralized extract had resolution adequate to enable the identification of the main phosphate compounds. The spectral intensities were converted to concentrations. The lower detection limit with extensive signal averaging was 0.02 Mmol for the extract. The composition, listed in order of decreasing concentration, … Show more

“…The most prominent peaks are due to phosphocholine, inorganic phosphate (P i ), the ␥-phosphate of nucleoside triphosphate (␥-NTP) (which also includes ␤-NDP), ␣-NTP (which also includes ␣-NDP), ␤-NTP, and uridine diphosphate sugars (UDPS). The assignment of the peaks was based on known chemical shifts of phosphate metabolites from previous studies (17,18), and by spiking the NMR spectra of uninfected HeLa cell extracts with uridine 5Ј-diphospho-N-acetylglucosamine, uridine 5Ј-diphosphoglucoronic acid, and uridine 5Ј-diphosphoglucose. A typical extract spectrum is shown in Fig.…”

“…Distinct from earlier 31 P MR spectroscopy (MRS) studies that reported total NAD contents (18)(19)(20), our approach is able to resolve the MR signal of NADH from that of NAD + by taking advantage of their specific spectroscopic characteristics at a given magnetic field strength that can be precisely predicted based on a theoretical model (17). Thus, both NAD + and NADH can be quantified simultaneously by matching the in vivo NAD spectra with the model-simulated spectra.…”

“…Expanding the spectra revealed another triphosphate near the P resonances of ATP (11) that accounted for 25 ± 4% (s.d., n = 3) of the total peak area in GLC4/ADR extracts and for 19 ± 4% in GLC4 extracts. This triphosphate could be UTP, GTP or CTP (Evans & Kaplan, 1977;Evanochko et al, 1984). High resolution 'H NMR spectra were obtained from extracts of both cell lines in D20.…”

“…Assignments were made on basis of data in the literature (Daly et al, 1987;Evans & Kaplan, 1977;Evanochko et al, 1984) and by adding standard compounds. Extracts of both cell lines showed high levels of PC (1 in Figure la and b).…”
