13C‐nmr spectroscopic investigation of two yeast cell wall β‐D‐glucans

Abstract: SynopsisBy means of alkaline extraction of the cell walls of the yeasts Saccharomyces cermisiue and Candida albicans, water-insoluble glucans were obtained. Methylation analysis and 13C-nmr investigation in dimethyl sulfoxide solution revealed the similar chemical structure of these glucans, being composed of p(1 + 3) glycosidically linked D-glucopyranmyl units with a small amount of p(1 4 6) linkages. More detailed study in dilute alkaline solutions and in the gel state a t neutral pH, however, showed that an… Show more

“…The disordered conformation of the L-FV-IA in DMSO is also similar to schizophyllan and Lentinan in 0.38 M NaOH aqueous solution. 10 15 and Schzophyllan in 0.06 M NaOD/D 2 O. 10 Moreover, with an increase of water content the relative intensities of C6 signal at 61 ppm significantly decreased and shifted to downfield by 1.2 ppm.…”

“…14 The gel-forming ability of polysaccharide is of considerable importance in view of their biological functions. 15 Moreover, helixcoil transitions are important in biology, as they appear in proteins and DNA. 16 In this work, the conformation transition for the β-(1 → 3)-D-glucan (Lentinan) from Lentinus edodes in water/DMSO mixture solution was investigated by laser light scattering, size exclusion chromatography, viscometry and 13 C NMR.…”

“…2B). The spectrum is characteristic of a strictly linear (133)-␤-D-glucan for which resonance peaks are expected at 61.0, 68.5, 72.9, 76.4, 86.3, and 103.1 ppm for C6, C4, C2, C5, C3, and C1, respectively (33,54,62). It is comparable to those obtained for wellcharacterized (133)-␤-D-glucans (33,62) and for in vitro (133)-␤-D-glucans synthesized by enzymes from S. monoica (54) and plants (14,36,37).…”

“…Fungal glucan was isolated from the cell wall of Saccharomyces cerevisiae as previously described (Kogan et al 1988, Chorvatovičová et al 1993. Candidate antagonistic Trichoderma strains from the collection of ISPA, Bari, Italy were tested against pathogenic Fusarium species isolated from maize ears and roots in dual cultures grown on potato-dextrose-agar (Sigma) for 7-21 days.…”
