Sumoylation of Peroxisome Proliferator-Activated Receptor γ by Apoptotic Cells Prevents Lipopolysaccharide-Induced NCoR Removal from κB Binding Sites Mediating Transrepression of Proinflammatory Cytokines

Abstract: R ecognition of apoptotic cells (AC)3 elicits immunological consequences that have received considerable attention during recent years. Professional phagocytes such as dendritic cells and macrophages recognize AC via so called "eat me" signals, with concomitant phagocytosis (1). The uptake of AC avoids secondary necrosis and thus the release of harmful cell contents. Moreover, ingestion of apoptotic material actively provokes a macrophage phenotype shift, which helps to terminate perpetuating inflammatory resp… Show more

“…When the oilseeds were added to the high-fat diet, they restored the level of TNFa expression down to the level found in mice fed the low-fat (10 %) diet, indicating an efficient amelioration of the diet-induced inflammation. Activated ERs, PPARs and LXRs are efficient inhibitors of NF-kB [62][63][64], suggesting that the antiinflammatory activity of lignans (as evidenced in Fig. 5) might also involve these NRs.…”

“…PIAS1 was initially named for its ability to interact with and inhibit signal transducer and activator of transcription (STAT) 1 (18), but subsequent investigations have clarified that PIAS1 regulates a variety of transcription factors through distinct mechanisms, including promoting SUMOylation of target proteins and blocking the DNA-binding activity of transcription factors (19). PIAS1 plays a critical role in such cellular processes as cell proliferation (20), cell differentiation (21, 22), and immune responses (23)(24)(25). In the innate immune system, PIAS1 occupies the binding sites of STAT 1 and NF-kB on the promoters of their target genes to block expression of proinflammatory factors such as TNF-a and macrophage inflammatory protein 2 (MIP2).…”

“…Surprisingly, the diminished inhibitory effect of oxLDLexposed cells on macrophage cytokine expression is not paralleled by changes in PPAR ␥ reporter activation. Previously, we showed an important role of PPAR ␥ in macrophage polarization in response to apoptotic cells regarding both oxidative burst and cytokine production ( 3,36 ). These discrepancies may refl ect different mechaby guest, on March 24, 2019 www.jlr.org Downloaded from nisms governing PPAR ␥ activation and suppression of gene expression or suggest PPAR ␥ -independent pathways accounting for a proinfl ammatory character of oxLDLtreated cells.…”
