Sulforaphane absorption and histone deacetylase activity following single dosing of broccoli sprout supplement in normal dogs

Abstract: The role of epigenetic alterations during cancer has gained increasing attention and has resulted in a paradigm shift in our understanding of mechanisms leading to cancer susceptibility. Sulforaphane (SFN) is a naturally occurring isothiocyanate derived from the precursor glucosinolate, glucoraphanin (GFN), which is found in cruciferous vegetables such as broccoli. Sulforaphane has been shown to suppress tumour growth by several mechanisms including inhibiting histone deacetylases. The objective of this study … Show more

“…Although SFN is water-soluble and well absorbed after oral administration both in humans and animals [ 25 , 26 ], the SFN bioavailability from cruciferous vegetables varies widely based upon many factors, mainly stemming from the conversion rate of GR to SNF by the enzyme myrosinase. This plant enzyme is commonly segregated from the glucosinolates until the cells are disrupted and is also produced by gut microbiome [ 27 ].…”

“…Dietary intake of SFN in the form of BroccoMax TM to the ten healthy dogs was peaked after 4 h from plasma concentration of SFN and SFN metabolites. In the canine patients uptake of SFN could have inhibited histone deacetylase (HDAC) activity [ 42 ]. It is also debatable that as GSLs hydrolyzed by gastrointestinal microbes, consumption of broccoli significantly reduced the gastrointestinal microbiota [ 43 ].…”

“…Among them include, phenyl ITCs that inhibit transcriptional activity regulated by androgen receptor modulating growth of prostate cancer cells through down-regulation of an androgen receptor-regulated gene [ 134 ]. ITCs may influence changes and activity of compounds that are normally overexpressed during cancer such as histone deacetylase [ 112 , 114 ] or compounds which may cause intoxication such as doxorubicin [ 124 ].…”
