Germination of cereals is accompanied by extensive change in the redox state of seed proteins. Proteins present in oxidized form in dry seeds are converted to the reduced state following imbibition. Thioredoxin (Trx) appears to play a role in this transition in cereals. It is not known, however, whether Trx-linked redox changes are restricted to cereals or whether they take place more broadly in germinating seeds. To gain information on this point, we have investigated a model legume, Medicago truncatula. Two complementary gel-based proteomic approaches were followed to identify Trx targets in seeds: Proteins were (1) labeled with a thiol-specific probe, monobromobimane (mBBr), following in vitro reduction by an NADP/Trx system, or (2) isolated on a mutant Trx affinity column. Altogether, 111 Trx-linked proteins were identified with few differences between axes and cotyledons. Fifty nine were new, 34 found previously in cereal or peanut seeds, and 18 in other plants or photosynthetic organisms. In parallel, the redox state of proteins assessed in germinating seeds using mBBr revealed that a substantial number of proteins that are oxidized or partly reduced in dry seeds became more reduced upon germination. The patterns were similar for proteins reduced in vivo during germination or in vitro by Trx. In contrast, glutathione and glutaredoxin were less effective as reductants in vitro. Overall, more than half of the potential targets identified with the mBBr labeling procedure were reduced during germination. The results provide evidence that Trx functions in the germination of seeds of dicotyledons as well as monocotyledons.

Water deficits cause large yield losses in wheat. Although anthesis is generally considered the most vulnerable period, water deficit during grain filling can also cause yield losses. The objective of this study was to investigate the effect of water stress and rewatering, at three different grain developmental stages, on physiological and grain filling parameters and on yield components. Wheat plants were subjected to water deficit and rewatering at the watery ripe, milk and soft dough stages. In the flag leaf, water stress decreased the relative water content, the chlorophyll and protein content and increased the leakage of solutes, at all three studied grain filling stages. Water stress at the watery ripe and milk stages reduced the final grain dry mass by 47 % and 20 %, respectively. This reduction was due to a decrease in the grain filling period and to a significant reduction in the maximum rate of grain-fill. Water stress imposed at the watery ripe stage reduced not only the linear growth phase but also its slope; grain number per spike and the 1000-kernel weight were also significantly reduced. SDS-PAGE patterns of grain proteins at the watery ripe stage did not differ between the controls, stressed or rewatered treatments. Protein patterns at the milk stage changed substantially with water stress, mainly for the high molecular weight glutenin subunits and ω gliadins. Three new bands were observed with apparent molecular weights of 108.5 kDa, 84.8 kDa and 63 kDa. Rewatering reverted water stress effects when it was imposed at the milk stage. Water deficit at the soft dough stage did not have any effect on protein grain patterns. Apesar da antese ser considerada, geralmente, a fase mais vulnerável, o déficit hídri-co durante todo o período de preenchimento também pode provocar perdas na produtividade da cultura. O objetivo do presente estudo foi pesquisar o efeito do estresse hídrico e da reidratação durante três estágios, no desenvolvimento do grão de trigo, sob diferentes parâmetros fisiológicos e do preenchimento dos grãos, e sob os componentes do rendimento. As plantas foram submetidas ao estresse hídrico e reidratação durante as fases de grão aquoso, leitoso e pastoso. O estresse hídri-co reduziu o conteúdo relativo d' água, o conteúdo da clorofila e das proteínas nas folhas bandeiras e aumentou a liberação dos solutos, nos três estádios do grão estudados. O estresse produzido durante os estádios aquoso e leitoso reduziu o peso final dos grãos em 47 % e 20 %, respectivamente. ABSTRACT. Thioredoxin h (Trxh) is a ubiquitous protein that reduces disulfides in target proteins, and is itself reduced by NADPHthioredoxin reductase. In the current study, the complementary DNA sequence and the genomic sequence of the three-pistil (TP) line of common wheat (Triticum aestivum L.) were obtained from spikes through reverse transcription-polymerase chain reaction (RT-PCR) and touchdown-PCR. Sequence alignment of amino acids of TPTrxh then allowed for predictions of its physicochemical properties, secondary structures, tertiary structures, and functional domains. Furthermore, the TPTrxh gene was overexpressed in Escherichia coli and its activity was demonstrated using a dithiothreitol-dependent insulin assay. The expression patterns of TPTrxh were analyzed through real-time RT-PCR in different tissues and across different developmental stages of young spikes. The complementary DNA of TPTrxh was found to be 411 bp in length, encoding 118 amino acids. Its genomic sequence was determined to be 2632 bp, possessing 3 exons and 2 introns. Functional domain analysis indicated that TPTrxh contained a WCGPC motif located at the end of the second β-fold and on the initial side of the second α-helix. The TPTrxh protein reduces intramolecular and intermolecular Cloning and characterization of thioredoxin h in TP lines disulfide bridges in target proteins. Young spikes contain higher levels of TPTrxh transcripts than do stems and leaves. The transcript levels in the young spikes (2-5 mm in length) of the Chinese Spring TP line increased 2.84-fold relative to those of young spikes (2-5 mm in length) of the Chinese Spring line. These data provide a basis for future research into the function of Trxh, and offer further insight into the molecular mechanism of the TP mutation in wheat.
