Sulfated Muco-Substances and Gastric Diseases

³⁵S-sulfate was incorporated into acidic mucopolysaccharides, various sulfated glycoprotein fractions and sulfatides during in vitro incubation of minced rat stomach (mucosa and muscle) in minimal essential medium containing ³⁵SO⁼₄. The labelled components were elaborated into the medium in addition to being present in the tissue proper; more radioactive sulfate was incorporated into the soluble or medium components than into the tissue-bound components. Detergent fractionation and electrophoresis studies suggest the presence of sulfated polyanions not previously described for this tissue. Chondroitin sulfates were the sole compounds desulfated by gastric enzymes, and this action may account for the observation that chondroitin sulfates from stomach tissues appear to be undersulfated. Gastric enzymes also liberated organic sulfates from chondroitin sulfates and from sulfated glycoproteins.

The outputs of organic and inorganic components of saliva were compared in healthy subjects and in patients suffering from an evolutive duodenal ulcer. Under stimulation of the salivary and pharyngeal secretion by a collection tube positioned in the stomach, the volume of saliva and the hexosamines, sialic acid, sulfate, total proteins, potassium and free calcium outputs were lower in DU than in controls. The differences were not observed when the salivary secretion was collected in patients chewing Parafilm, when pharyngeal secretionwas not stimulated.

Sulfated Glycoprotein Biosynthesis in Human Gastric Mucosal Biopsies

Sulfated mucus glycoprotein biosynthesis by human gastric mucosal biopsies was studied. Human gastric healthy specimens and cancer specimens were obtained from antral and corpus normal mucosal areas by biopsy. A comparison was made of the biosynthesized sulfated mucus glycoproteins assessed by the incorporation of ³⁵S-sulfate and ¹⁴C-glucosamine into the specimens using an organ culture technique. A significant uptake of ³⁵S-sulfate or ¹⁴C-glucosamine into the mucus glycoproteins was demonstrated in all specimens. ¹⁴C-glucosamine incorporation into the glycoproteins was almost the same for all biopsies. The synthesis of sulfated glycoprotein in healthy antral mucosa was about four times that in corpus mucosa. Significantly raised levels in sulfated glycoprotein biosynthesis were found in cancer specimens, being about 30 and 13 times higher in antrum and corpus mucosa than that of healthy specimens, respectively.
