The tepals of cut tulips (Tulipa gesneriana L. cv. Ile de France) kept at 20 °C had severely wilted 7 days after flower opening. Suppression of abscission and undesirable growth of tepals is required to extend vase life. Treatment with 50 mm trehalose in combination with 50 μm chloramphenicol (CAP) delayed abscission by 4 days compared with stems placed in distilled water or CAP without trehalose. Only 4% of trehalose+CAP-treated flowers exhibited tepal abscission 7 days after harvest, while 82% and 60% of flowers held in distilled water and CAP, respectively, did so; the tepals of trehalose+CAP-treated flower stems contained 50% more water than did those treated with CAP alone. Further, trehalose did not promote elongation of epidermal parenchyma cells in tepal tissues, but maintained radial enlargement of the cells. Thus, trehalose+CAP treatment is effective in prolonging vase life without abscission, water loss, or elongation of cells in tulip tepals, but slight wilting occurs in leaves.

Vase life of ‘Karma Thalia’ dahlia (Dahlia ×hybrida), ‘Lace Violet’ linaria (Linaria maroccana), ‘Sunrise’ lupine (Lupinus hartwegii ssp. cruickshankii), ‘Temptress’ poppy (Papaver nudicaule), ‘Indian Summer’ rudbeckia (Rudbeckia ×hybrida), ‘Jemmy Royal Purple’ trachelium (Trachelium caeruleum), and ‘Benary's Giant Scarlet’ and ‘Sun Gold’ zinnias (Zinnia elegans) was determined after being subjected to postharvest handling procedures. Cut dahlia, lupine, poppy, rudbeckia, trachelium, and ‘Sun Gold’ and ‘Benary's Giant Scarlet’ zinnia flowers could be held in unamended tap or deionized (DI) water with no effect on vase life. Vase life of linaria was longest when placed in DI water with 8-hydroxyquinoline citrate and a solution pH of 3.5. A vase solution of 2% sucrose without foam extended consumer vase lives for linaria, trachelium, and ‘Benary's Giant Scarlet’ zinnia. Floral foam or 2% or 4% sucrose had no effect on the consumer vase life of dahlia, lupine, rudbeckia, and poppy. Trachelium and rudbeckia did not tolerate a 20% sucrose treatment for 24 h, whereas linaria and ‘Benary's Giant Scarlet’ zinnia had a longer vase life with a 10% sucrose pulse than a water-only pulse. For trachelium, the longest (17.5 days) consumer vase life occurred when the Chrysal Professional 2 Processing solution (CP2) was used after pretreatment with DI water. Either of two commercial holding solutions, CP2 or Floralife Professional (FLP), similarly extended the vase life of linaria. The use of FLP or CP2 improved consumer vase life of dahlia, lupine, and poppy compared with DI water. Dahlia, trachelium, and zinnia flowers could not be cold stored at 2 °C. Lupine and poppy could be stored at 2 °C wet or dry for 2 weeks. Linaria and rudbeckia could be cold stored for 3 weeks. Lupine and trachelium were susceptible to 1 μL·L−1 exogenous ethylene, which induced floret abscission in lupine and stopped floret opening in trachelium. 1-Methylcyclopropene and silver thiosulfate similarly suppressed the ethylene effect. Cut linaria, zinnia, dahlia, rudbeckia, and poppy flowers were unaffected by exogenous ethylene.

The purpose of this study was to clarify the effect of sucrose on auxin-induced growth of stem excised from growing tulips and excised directly from cooled and not cooled bulbs, and on the growth of excised IV internode from growing plants in the presence of auxin. In all cases flower bud was replaced by IAA (indole-3- acetic acid, 0.1%, w/w in lanolin) and basal part of excised segments of stem was kept in distilled water or in solution of various sugars at different concentrations. IAA-induced growth of excised stems isolated from growing tulips was inhibited by sucrose at concentrations of 5.0% and 10.0%, but sucrose at 1.25% and 2.5% did not. Sucrose at all concentrations used evidently delayed senescence and increased chlorophyll contents in excised stems in the presence of IAA. Sucrose induced stiffing in isolated stems in the presence of IAA, and much less infective by pathogen in comparison to stem treated with IAA only. Mannitol and sorbitol at concentrations of 5.0% and 10.0% substantially inhibited IAA-induced growth of stem segments. Stem segments excised from cooled and not cooled tulip bulbs were more sensitive than those isolated from growing shoots due to application of sucrose and glucose; more inhibitory effect was observed. Sucrose at concentrations of 5.0% and 10.0% only slightly inhibited growth of IV internode treated with IAA and all concentrations of sucrose (1.25%, 2.5%, 5.0% and 10.0%) substantially increased chlorophyll content. The possible mode of actions of sucrose interacting with auxin to regulate stem growth is also discussed although sugar response is complicated by the fact that plants have multiple sugar-response pathways.
