Successive Construction of Cellulase Hyperproducers of Trichoderma Using Hyperpolyploids

Abstract: When the swollen conidia of Trichoderma reesei QM 6a are treated with 0.1% (w/v) colchicine solution, huge autopolyploid nuclei can be formed in those swollen conidia. When a mycelial mat derived from such a conidum is treated with a haploidizing reagent, benomyl, many fan-shaped sectors are produced from the colony, and cellulase hyperproducers are selected from conidia on the colony. When colchicine and benomyl treatments are repeated on cellulase hyperproducers, new hyperproducers can be constructed success… Show more

“…Colchicine has been shown to induce polyploidy in plants by inhibiting mitosis (2,3). Colchicine has also been shown to induce polyploidy in fungi and Basidiomycetes (4,5). When fungal conidia are incubated in a liquid medium containing colchicine, the diameter of nuclei in conidia gradually increase and generate polyploid nuclei.…”

“…For this purpose, a system must be developed to rapidly isolate cellulase hyperproducers of this fungus. We earlier formed autopolyploids of this fungus using polyploidizer with swollen conidium [5]. Moreover, we could develop a double-layer selection medium that can rapidly isolate the strains with higher degrading ability of crystalline cellulose [6].…”
