Successful Treatment of Infantile Hemangiomas with Interferon-α-2b

Abstract: Interferon-alpha-2b therapy is an effective, well-tolerated treatment for complicated hemangiomas. Measurement of urinary bFGF levels may provide an objective method for monitoring treatment response.

“…The transcriptional effects of these cytokines on EC, however, have been only partially addressed (31) despite the broad evidence for their antiangiogenic effects (32)(33)(34). The aim of this work was to compare the in vitro effects of type I and type II IFNs on the transcriptional profile of EC of different origins and sources.…”

“…However, side effects such as delayed skeletal growth, depressed immune function, and personality changes limit their usefulness, particularly when long-term treatment is required. In lifethreatening cases that have not responded to corticosteroids, IFN-␣ has been used as an alternative (3,4). IFN-␣ is believed to inhibit proliferation of endothelial cells; however, its use has been limited by reports of severe and irreversible neurotoxicity in as many as 20% of patients treated (5,6).…”

“…Children with endangering or lifethreatening hemangiomas are treated with corticosteroid or interferon-␣. 3,4 However, not all hemangiomas respond to these drugs and neurological toxicity has been associated with interferon-␣ administration. 5,6 The recent demonstration of ectopic expression of type 3 iodothyronine deiodinase in hemangioma, which in at least one child caused hypothyroidism, 7 underscores the need for novel therapy.…”
