Successful fertility following optimized perfusion and cryopreservation of whole ovary and allotransplantation in a premature ovarian insufficiency rat model

Abstract: BackgroundFertility preservation by whole ovary cryopreservation and transplantation (WOCP&TP) with vascular anastomosis requires successful cryopreservation. In this study, we investigated the possibility of restoring ovarian function and natural fertility after WOCP&TP in a premature ovarian insufficiency (POI) rat model. The influence of cryopreservation on the offspring of rats following WOCP&TP was also explored.MethodRats aged 8-10 weeks were used as donors and recipients for allotransplantation. Fifteen… Show more

“…Vascular transplantation of fresh whole ovaries has been largely successful in animal models (rats [ 103 , 104 , 105 ], rabbits [ 106 , 107 , 108 , 109 , 110 , 111 , 112 , 113 , 114 ], sheep [ 47 , 49 , 67 , 68 , 78 , 115 , 116 , 117 , 118 , 119 , 120 , 121 ], pigs [ 122 , 123 ], dogs [ 124 ] and monkeys [ 125 , 126 ]). Encouraging results have also been reported on transplantation of frozen–thawed whole ovaries in rats [ 66 , 127 , 128 , 129 ], rabbits [ 130 ] and sheep [ 38 , 40 , 41 , 48 , 49 , 50 , 67 , 68 , 78 , 95 , 102 , 131 ], which will be detailed further in this review. In humans, however, only fresh whole ovary transplantation has been undertaken so far [ 51 , 132 , 133 , 134 ], yielding one live birth to date [ 51 ].…”

“…Recently, the efficacy of whole ovary cryopreservation and transplantation was investigated in a POI rat model, simulating female patients with cancer treated by chemotherapy [ 129 ]. POI was induced in 25 recipient rats by administering cyclophosphamide and evidenced by rising serum FSH levels and decreasing progesterone and estradiol values.…”

“…There are two UTx groups in Shanghai. Hua and Liu’s group at the Obstetrics and Gynecology Hospital (Fudan University) has done extensive research on preservation of female fertility [24,25]. This group has treated a significant number of patients with genital malformation who wanted to have their own child, and are committed to extending UTx to the clinical stage [26,27].…”

“…As many studies have confirmed, the influence of the freezing time on cell viability is negligible. In our previous studies, short-term cryopreserved ovaries [ 8 , 9 ] and bladder mucosa [ 10 ] survived in the transplanted subjects. Moreover, various tissues, such as cartilage [ 11 ] and heart valves [ 12 , 13 ], have been successfully cryopreserved and banked for more than 6 months and achieved transplantation effects that were equivalent to those of fresh controls.…”
