Successful Establishment of in Vitro Cultures of Prunus Cerasifera Hybrids by Embryo Culture of Immature Fruits

Abstract: Rapid obtaining of clonal plants is desirable to shorten crossing programs in fruit tree breeding. In this work, seedling germination and multiplication in vitro allowed us to establish successful in vitro cultures of hybrid seedlings. As a part of a breeding program in apricot rootstocks, we have obtained interespecific hybrid seeds after cross-pollination: myrobalan x apricot (Prunus cerasifera  armeniaca). Immature fruits were harvested and embryos, isolated in aseptic conditions, were grown in culture med… Show more

“…A significant percentage of embryos were smaller than 8 mm at harvest, what is considered as abortive seeds (Arbeloa et al, 2000). In optimal in vitro germination conditions, germination rate was 95.6% for embryos larger than 8 mm (non-abortive seeds) and 60% for the embryos smaller than 8 mm (abortive seeds) (Daorden at al., 2002;Arbeloa et al, 2003;Daorden et al 2004). Moreover, whereas in vitro multiplication of Myrobalan is easy and rapid (Hammerschlag, 1982) apricot is considered a recalcitrant species (Marino et al, 1993).…”

“…Germination and survival rates were recorded for each treatment. Shoots were later rooted in vitro in half MS medium with 5M IBA (Arbeloa et al, 2003). Micropropagated plants were acclimatized (Marín, 2003) and finally transplanted to orchard conditions.…”

“…Overcoming post-zygotic barriers and saving at least some hybrid embryos is possible through the use of the embryo-rescue technique (Golis et al 2002;Kukharchyk, Kastrickaya 2006;Liu et al 2007). It enables the development and growth of immature embryos in vitro, which makes it a very useful tool in the distant hybridization of plants of the genus Prunus (Arbeola et al 2003).…”

“…However, the plant regeneration rate was still not high enough and should be increased with more measures. In several species of Prunus, immature embryos were usually kept at a low temperature (0-4°C) more than 2 months to avoid the development of rosette type plants (Arbeloa et al, 2009;Burgos and Ledbetter, 1993;Kukharchyk and Kastrickaya, 2006). We noticed the formation of rosette-type structure without shoot tips from embryos which accounted for about 30% of the total cultured embryo in 2012.…”
