Subunit II of cytochrome c oxidase from Paracoccus denitrificans

Abstract: Cytochrome c oxidase from the bacterium Paracoccus denitrcjicans, while being related to the mitochondrial enzyme in many ways, consists of only two to three different subunits. For the identification of its genes, a Paracoccus DNA library was constructed and screened with specific antibodies for expression of cloned inserts in E. coli. A positive clone expressing immunoreactive products in the molecular mass region of authentic subunit I1 revealed a high homology of its DNA-deduced amino acid sequence with su… Show more

“…Expression and Purification Procedures-Expression of the Cu A fragment of P. denitrificans subunit II, encoded by the ctaC gene (20), was carried out in Escherichia coli JM 109. The region representing amino acid residues 130 -280 of the Cu A domain was amplified by PCR.…”

“…FliP has a cleavable signal peptide at its N-terminus (Ohnishi et al, 1997), an unusual modification of prokaryotic cytoplasmic membrane proteins (e.g. Sugimoto et al, 1977;Steinrucke et al, 1987).…”

“…These hydrophobic sequences are separated by hydrophilic residues that frequently contain clustered positive charges. In Chou-Fasman predictions, the internal hydrophobic sequences are predicted to be alpha- Computer-assisted searches of protein sequence data banks showed no significant similarity to any sequence contained in these banks and, in particular, no similiarity to the highly conserved bacterial and mitochondrial cytochrome aa3 polypeptides (37,42,45). This finding is also consistent with the isolation of a compensatory (bypass) mutation in a ctaA null mutant that restores cytochrome aa3 (Table 3) and confirms the inference that ctaA does not encode a cytochrome aa3 subunit.…”

“…Although Paracoccus denitrificans has provided the paradigm for biochemical and biophysical analysis of bacterial cytochrome aa3 complexes (26), the lack of a well-defined genetic system hinders analysis of cytochrome aa3-deficient mutants and the cloned subunit genes of this bacterium (37,45). However, the array of genetic manipulations available in precludes such studies in this.…”
