Substrate profiling of Zika virus NS2B‐NS3 protease

Abstract: Zika virus (ZIKV), isolated from macaques in Uganda in 1947, was not considered to be a dangerous human pathogen. However, this view has recently changed as ZIKV infections are now associated with serious pathological disorders including microcephaly and Guillain-Barre syndrome. Similar to other viruses in the Flaviviridae family, ZIKV expresses the serine protease NS3 which is responsible for viral protein processing and replication. Herein, we report the expression of an active NS3 pro domain fused with the … Show more

“…Proteases containing a glycine linker between NS2B CF and NS3 pro are denoted as 'linked', even when they undergo autocatalytic cleavage during purification. Kinetic data are derived from a protease assay (25°C) employing a fluorogenic tripeptide (Boc-GRR-AMC) [20], melting temperatures are based on tryptophan fluorescence measurements ( Figs 2B and S2C). For detailed information see Table S1.…”

“…Here, we mutated these sites individually and thus identified the relevant autocatalytic cleavage site to be at position R95 NS2B in the C‐terminus of the NS2B CF . This finding is intriguing for multiple reasons: The ZIKV NS2B CF ‐NS3 pro is believed to be specific toward dibasic residues . However, in the frequently used ZIKV NS2B CF ‐NS3 pro construct, cleavage occurred at a single basic residue R95 NS2B which is preceded by a methionine.…”

“…This might strongly influence how side chains, specially at the P4 position, bind into the different active site pockets. Indeed, using different capping group in PS-CSL of substrates has been shown to result in different amino acid preferences in the S4 pocket of the Zika virus NS2B-NS3 protease [53,54]. Also, if cooperativity exist between the P4 and other positions, the influence of the capping group on the S4 pocket specificity will also influence that of other pockets.…”

“…Regarding NS2A, to our knowledge, there are no specific studies addressing the function of this protein in ZIKV. On the other hand, NS3 is a trypsin-like serine protease involved in polyprotein processing (Gruba et al, 2016;Lei et al, 2016) that also exhibits helicase activity, which plays a pivotal role in viral RNA replication enabling RNA unwinding . Apart from its involvement in viral polyprotein processing, NS3 can cleave cellular factors such as FAM134B, hence modulating the autophagic response within ZIKV-infected cells (Lennemann and Coyne, 2017).…”
