Subpicosecond resonance Raman spectra of the early intermediates in the photocycle of bacteriorhodopsin

Abstract: The resonance Raman spectra are presented for the species formed during the photocycle of bacteriorhodopsin (bR) on a timescale of 800-900 fs. In the ethylenic stretch region two intermediates were found with frequencies of 1,510 and 1,518 cm(-1), corresponding to species with optical absorption maxima at 660 and 625 nm, respectively. This leads to the assignment of the 1,518 cm(-1) band to the J(625) intermediate. In the fingerprint region, the appearance of a vibration at 1,195 cm(-1) strongly suggests that … Show more

“…However, a number of experimental observations (24)(25)(26); see references cited in ref. 23 for earlier dynamics) unambiguously demonstrated the flat character of the excited state surface and the presence of a small barrier. Therefore, the ''three-state'' model (24,27,28), proposed that both the S 1 (B u ) and S 2 (A g ) states are involved in the dynamics of the excited state photoisomerization.…”

“…1) is not exponential and can be fitted to two lifetime components of Ϸ500 fs (80-95%) and 3 ps (20-5%) (16)(17)(18)(19)(20)(21)(22)(23). With a better signal-to-noise ratio, the early lifetime itself is found to be biexponential with 0.24 and 0.75 ps components in the ratio of 2:1 (24).…”

“…These observations are best explained by a small conformational relaxation of the chromophore and its immediate protein environment that occurs as the photoproduct evolves from a distorted form to a relaxed all-trans geometry (6,7,21). Conformational changes in both rhodopsin and bacteriorhodopsin on time scales >700 fs have been observed by time-resolved vibrational techniques (5,20,22).…”

“…Time-resolved resonance Raman (RR) spectroscopy has been used to determine changes in the vibrational degrees of freedom associated with these early intermediates (2,(15)(16)(17)(18)(19)(20). Although these studies have yet to firmly characterize the structural nature of J-625 (16,18,20), it appears that J-625 contains a highly twisted retinal [deduced from the strong HOOP modes observed in the 900-to 1000-cm-1 region (16,18)].…”
