Submicromolar concentrations of 4-hydroxynonenal induce glutamate cysteine ligase expression in HBE1 cells

Abstract: Abstract4-Hydroxynonenal (HNE), a major electrophilic product of lipid peroxidation, is regarded as both a marker of oxidative stress and a mediator of oxidative damage. At subtoxic concentrations, however, this compound has been shown to be a signalling molecule that can induce the expression of various antioxidant/detoxification enzymes, including glutamate-cysteine ligase (GCL), the rate-limiting enzyme in the de novo synthesis of glutathione. GCL consists of a catalytic (GCLC) and modulatory (GCLM) subunit… Show more

“…Our observations are consistent with prior reports that Nrf2 played a critical role in regulating expression of GCLc and GCLm. 67,68 Our results suggest that the elevation of intracellular redox status might be one of the mechanisms by which Plin5 inhibits HSC activation.…”

“…As low as 0.5 μM, HNE can induce transcription of glutamate cysteine ligase, the first enzyme of GSH synthesis, through activation of the Nrf2-EpRE signaling pathways (198). All these studies point to the potential signaling function of HNE in both physiological and pathological conditions.…”

“…Stressdependent induction of Nrf2 signaling directly regulates the transcription of several antioxidant enzymes, including glutamate cysteine ligase (GCL; both GCLC and GCLM subunits), 25 glutathione S-transferase (GST ), 17 glutathione peroxidase (GPx), 17 glutathione reductase (GR), heme oxygenase-1 (HMOX1), and NADP(H) quinone oxidoreductase 1 (NQO1). GCL, GST, GPx, and GR are involved in glutathione (GSH) biosynthesis and its recycling.…”
