Subcloning and Restriction Enzyme Mapping of Bovine Adenovirus Type 2

Abstract: Bovine adenoviruses (BAVs), causing both respiratory and/or enteral diseases in cattle, have been isolated in many countries all over the world. In this study we report on the molecular cloning of the internal EcoRl fragments spanning 20.6-90.5%, and the internal Sall fragments spanning 3.1-65.2% of the BAV type 2 (BAV2; strain No. 19) genome into the plasmid vector pUC19. Moreover, the subcloning of the BAV2 genome facilitated the construction of a detailed physical restriction endonuclease map for BamHl, Cla… Show more

“…Bovine adenoviruses type 2 (BAV2) and bovine adenovi ruses type 3 (BAV3) in subgroup 1 are among the best characterized BAVs. In general, their genomic organiza tion is similar to those of other Ads [2,3] This article is also accessible online at: ww\v. karger.com http://BioMcdNet.com/karger host range, however, BAV2 and BAV3 differ from each other.…”

“…We have previously reported the construction of a library of plasmids containing restriction fragments of the BAV2 strain 19 [8] and BAV3 strain WBR-1 [7] viral genomes cloned in pUC19. For BAV2, three plasmids containing 0-17.5% (pBB12), 5.1-19.3% (pPE30) and 19.3-30.6% (pEB113) of the viral genome were used for sequencing.…”

“…Our previous analysis of the BAV2 and BAV3 genomes revealed that their genetic organizations are similar to other Ads belonging to the Mastadenovirus subfamily [7][8][9][10]12]. Since all known sequences for Ad pol indicate that the gene is located between 10 and 24% of the genome, we targeted similar regions in the DNA of BAV2 and BAV3 for sequencing analysis.…”

“…The size of the genome reported varies from 30.3 to 34.8 kb [9][10][11][12] and restriction maps of BAV2 (No. 19) for a num ber of enzymes are available [ 10,12], The main difference …”
