Subclassification of the presynaptic α2‐autoreceptors in rabbit brain cortex

Abstract: 1 a2-Adrenoceptor binding sites have been subclassified into a2A sites of which a main characteristic is very low affinity for prazosin, and a2B sites with relatively high affinity for prazosin. The presynaptic a2-autoreceptors in rabbit brain cortex were studied in order to classify them in terms of a2A and a2B* Release of [3H]-noradrenaline in cortical slices was elicited by trains of 4 pulses delivered at 100 Hz. 2 Clonidine caused concentration-dependent inhibition of the stimulation-evoked overflow of tri… Show more

“…Four studies, to our knowledge, have addressed the question directly, measuring the release of [3H]-noradrenaline or postsynaptic responses to neural stimulation. In contrast to the suggestions mentioned, the autoreceptors were classified as M2A in rat vas deferens (Connaughton & Docherty, 1990), rat brain cortex (Gobbi et al, 1990), rabbit brain cortex (Limberger et al, 1991) and guinea-pig urethra (Alberts, 1992); only in rat atrium the autoreceptors seemed to be M2B (Connaughton & Docherty, 1990). However, the classification of the rat atrial receptors as O2B, i.e.…”

“…M2A-Adrenoceptors are reported to be present prejunctionally in the rat vas deferens (Connaughton & Docherty, 1990), rat submandibular gland , and rabbit brain cortex (Limberger et al, 1991). The low potency of prazosin prejunctionally at the cholinergic nerves of guinea-pig ileum (Kaposci et al, 1987), at the noradrenergic and 5-hydroxytryptaminergic nerves of rat brain synaptosomes (Gobbi et al, 1991), and at antagonizing actions of endogenous noradrenaline in rat cerebral cortex (Harsing & Vizi, 1991), may suggest that these receptors are also x2A.…”

“…synaptic a2-autoreceptors were compared in rat heart and vas deferens (Doxey & Everitt, 1977). In rabbit brain cortex, pulmonary artery, kidney and atria the a2-autoreceptors have been characterized as a2A (Limberger et al, 1991;1995a;Molderings & G6thert, 1992;Trendelenburg et al, 1993). Based on these results, it has been proposed that 'the majority of a2-autoreceptors generally are a2D in the rat and a2A in the rabbit.…”
