Subcellular localization of m2 muscarinic receptors in GABAergic interneurons of the olfactory bulb

Abstract: We analysed the ultrastructural distribution of the m2 muscarinic receptor (m2R) in the rat olfactory bulb (OB) using immunohistochemical techniques and light and electron microscopy. m2R was differentially distributed within the cellular compartments of gamma-aminobutyric acid (GABA)ergic bulbar interneurons. It is located in the gemmules of granule cells and in the synaptic loci of the interneurons of the external plexiform layer, suggesting that m2R activation could modulate the release of GABA from these i… Show more

“…Consistently, bulbar neurons express nicotinic-type and muscarinic-type cholinergic receptors (Castillo et al, 1999;Crespo et al, 2000). However, different groups have made conflicting observations of cholinergic effects on bulbar neurons.…”

“…Electron microscopy Four wild-type and five transgenic brains were prepared for EM analysis (Crespo et al, 2000) and electron micrographs were randomly taken from the stratum radiatum of the CA1 region at a final magnification of ×26,000. Vesicles were considered docked when their membrane was less than 50 nm from the presynaptic active zone (Rosahl et al, 1995).…”

“…Vesicles were considered docked when their membrane was less than 50 nm from the presynaptic active zone (Rosahl et al, 1995). Post-embedding GABA immunostaining followed the method described by Crespo et al (Crespo et al, 2000), using a rabbit anti-GABA antiserum and 10 nm colloidal gold-coated secondary antibodies (Sigma). Electron micrographs at final magnifications of ×52,000 were taken at random from the stratum radiatum.…”

“…The activity of the neuromodulatory systems changes with the behavioral state of the animal. Because granule cells in the OB express muscarinic acetylcholine receptors on their spines in the EPL (Crespo et al, 2000), we hypothesized that the state-dependent change in cholinergic input might regulate dendrodendritic synaptic transmission in the OB in a state-dependent manner.…”
