Study on the Interaction of Vitamin B12 with DNA by Spectroscopy and Electrochemical Methods

Abstract: The Interaction between vitamin B 12 (VB 12 ) and fish sperm DNA was investigated in physiological buffer (pH 7.4) using the methylene blue (MB) dye as a spectral probe by spetcrophotometery, viscosity measurements and cyclic voltammetry. The apparent binding constant of vitamin B 12 with DNA was found to be 3. The results indicated that vitamin B 12 can bind to DNA and the major binding mode was intercalative binding.

“…Where, A and A 0 are the absorbance intensity of 5‐DDMP complex with dsDNA and 5‐DDMP, respectively. K is the binding constant, ϵ G and ϵ H–G are the absorptivity coefficients of 5‐DDMP and 5‐DDMP‐dsDNA complex, respectively and [DNA] is the molar concentration of dsDNA [36, 37]. The binding constant value came out to be 2.15×10 3 L mol −1 calculated by dividing intercept with slope of plot of A 0 /( A − A 0 ) vs 1/[DNA].…”
