Study on Extraction, Physicochemical Properties, and Bacterio-Static Activity of Polysaccharides from Phellinus linteus

Abstract: We optimized an ultrasound-assisted extraction process of Phellinus linteus mycelium polysaccharides (PLPs) and studied their monosaccharide composition and bacteriostatic properties. Based on a single-factor experiment, a three-factor, three-level Box–Behnken design was used to optimize the ultrasound-assisted extraction process of PLP, using the yield of PLP as the index. The chemical composition and monosaccharide composition of PLP were determined by chemical analysis and HPLC analysis, respectively. Micro… Show more

“…The purified PHP-1 was homogeneous and its molecular weight was determined by high-performance gel permeation chromatography (HPGPC) analysis (Li et al, 2022;Zhang et al, 2021) to be 60.99 kD. And the monosaccharide composition of PHP-1 was determined by high-performance liquid chromatography (HPLC) analysis (Eom et al, 2021;Gao et al, 2023) as ribose, glucuronic acid, galacturonic acid, glucose, galactose, and arabinose at a molar percentage of 0.9: 2.5: 0.5: 93.1: 1: 0.9 (Pu et al, 2022). The DMEM (Dulbecco's modified Eagles medium) and FBS (fetal bovine serum) were purchased from Gibco (NY, USA).…”

“…A polysaccharide composed of glucose, galactose, mannose, fucose, and xylose in a molar ratio of 4.36: 2.34: 2.09: 0.88: 0.42 showed prebiotic potentials by increasing amounts of beneficial bacteria, such as Bacteroides , Prevotella , and Butyricimonas , and reducing pathogenic bacteria including Escherichia , Shigella , Morganella , and Intestinimonas [ 129 ]. Besides that, a polysaccharide with a monosaccharide composition of fucose, rhamnose, arabinose, glucuronic acid, galactose, glucose, and xylose (1.4:0.5:0.9:1.6:4.7:84.8:6.0) exerted bacterio-static activities against bacteria Staphylococcus aureus , Escherichia coli , and Bacillus subtilis , demonstrating the potential of T. linteus polysaccharides for clinical applications [ 130 ].…”
