This paper is focused on a combined experimental and theoretical study of 3-substituted coumarin derivative, the 2-oxo-2H-chromen-3-yl acetate (I). The compound was synthesized by reacting chroman-2,3-dione and acetic anhydride in dried diethyl ether in the presence of dried pyridine and crystallized in the orthorhombic crystal system with Pbca space group. The lattice parameters of the structure are a=14.6770 (1), b=7.1079 (1), c=17.6767 (2) Å, α=β=γ=90° with 8 molecules per unit cell (Z=8). The compound has been characterized structurally by Spectroscopy utilizing 1 H NMR, 13 C NMR and IR techniques and by crystallography using the X-Ray diffraction (XRD) analysis. In the crystallographic study, the positions of the atoms were determined by direct methods and refined to a final R value of 0.038 for 1768 independent reflections. The stabilization of the structure is provided by intermolecular C-H•••O hydrogen bonds extending along [010] direction. Likewise, the presence and nature of intermolecular contacts are determined by the 3-D molecular Hirshfeld surface and 2-D fingerprint plot analysis which indicate the main contributions to the Hirshfeld surface, 38.7% for O ••• H and 28.7% for H •• H. Moreover, the molecular geometry of (I) was as well minimized utilizing density functional theory (DFT/RB3LYP), the frequency calculations with RB3LYP method, the basic ab initio model i.e the restricted Hartree-Fock (RHF) and the exchange component of Perdew and Wang's 1991 functional B3PW91 methods with the 6-311 ++ G(d, p) basis set in ground state. The derived structural parameters highlight very good correlation with the crystallographic results. Frontier molecular orbitals (HOMO-LUMO), their energy gap, the non-linear optical effects (NLO) and related reactive parameters were also computed to better apprehend the properties of the molecule.

Contrast compounds, also called contrast agents or contrast media are substances that temporarily change the way X-rays or other imaging tools interact with the body. Iomeprol is a nonionic monomeric iodinated contrast medium. Optimized geometries of Iomeprol, HOMO-LUMO energy, energy gap, global chemical indices, total energy, nonlinear optical and Natural Bond Orbital (NBO) analysis in the gas phase and in solvents (chloroform, acetic acid, ethanol, DMF, DMSO and water) were obtained based on Density Functional Theory with B3LYP/lanl2dz basis set. The results revealed that the solvents have an effect on the optimized and some chemical parameters (hardness, electronegativity, electrophilicity index, nucleofugality,..). It was observed EHOMO values become more negative and ELUMO values become more positive due to the increase dielectrical constant of the solvent. It was observed that Iomeprol had a highest stability, harder and less reactive in the water phase. In the NBO analysis, interaction between donor and acceptor electrons was effected with the dielectric constant of the solvent.

Source of materialsThe title compound was synthesized from the Sonogashira Coupling-Aminovinylation Sequence reaction [4]. In a nitrogen atmosphere, a THF solution of phenylacetylene (1.1 mmol in 5 mL THF) was added dropwisely into a stirred mixture of 2-bromo-5-nitrothiazole (1 mmol), Pd(PPh 3 ) 2 Cl 2 (14 mg, 0.02 mmol), CuI (7 mg, 0.04 mmol), and 11 mL of THF/NEt 3 (10:1). The reaction mixture was stirred at room temperature overnight until conversion was complete (monitored by TLC). Then, a solution of diethylamine (2 mmol) in 5 mL of methanol was added, and the mixture was heated to reflux for ca. 9 hour until complete conversion (monitored by TLC). The solvents were evaporated in vacuo, and the residue was purified by chromatography over a short pad of aluminum oxide eluting with dichloromethane followed by acetone to yield 60% of the entitled compound. The residue was then recrystallized in dichloromethane by evaporation in vacuo, which yielded dark red crystals. After several attempts a suitable crystal for single X-ray diffraction study was obtained. Experimental detailsH atoms were fixed geometrically at ideal positions and allowed to ride on the parent atoms with C-H = 0.93-0.97 Å, with U iso (H) = 1.5Ueq(C) (for CH 3 )and 1.2 Ueq(C) (for CH 2 and CH).
