Study of the possible effect of sacubitril/valsartan combination versus valsartan on the cognitive function in Alzheimer’s disease model in rats

Abstract: Objectives: Alzheimer’s disease (AD) is an irreversible, progressive neurodegenerative disorder. The proportion of elderly individuals at risk for AD and cardiovascular problems increases by raising life expectancy. The present study was designed to investigate the effect of the sacubitril/valsartan combination compared to that of valsartan alone in a rat model of AD. Methods: 72 male adult Wistar rats were divided into seven groups; control untreated rats received saline, control valsartan-treated rats receiv… Show more

“…Besides, NEP inhibitors increase the risk of developing late‐onset axonal polyneuropathy 41 . An updated experimental study demonstrated that LCZ696 increases AD risk compared with valsartan alone in rats 48 . Valsartan can decrease the risk of AD development as it selectively blocks AT 1 R as RAS inhibitors have been associated with the reduction of brain damage in different experimental and clinical models of neurodegenerative diseases 49 .…”

“…A recent study assessed the effects of sacubitril/valsartan (a relatively new antihypertensive combination) in comparison with valsartan alone on a rat model of AD (aluminum-induced). The results showed that valsartan alone improved AD symptoms and did not increase the risk of developing AD, while the sacubitril/valsartan combination worsened the condition of the treated animals and increased their risk of developing AD [ 107 ].…”
