Study of the correlation between fengycin promoter expression and its production by Bacillus subtilis under different culture conditions and the impact on surfactin production

Abstract: This work aimed to rely expression of the fengycin promoter to fengycin production under different culture conditions. To this end, Bacillus subtilis BBG208, derived from BBG21, which is a fengycin overproducing strain carrying the green fluorescent protein (GFP) under the control of fengycin promoter, was used to assess the effects of different carbon and nitrogen sources on surfactin and fengycin production and the fengycin promoter expression. The data showed that some carbon sources oriented synthesis of o… Show more

“…subtilis 168 was only 42 ± 7 μg/mL . A higher yield of 768 mg/L was obtained by optimizing the culture parameters combined with promoter replacement . The yield reached as high as 1600 mg/L upon disruption of the srf operon and completes blocking of the surfactin production through CRISPR/Cas9.…”

The procedure is easy to perform and provides an effective strategy for producing plipastatin at scale.…”

“…Therefore, we have attempted to improve plipastatin production by supply of seven amino acids of the plipastatin peptide chain in the mineral salt medium. Previously, these seven amino acids as the only nitrogen source were used in the mineral salt medium and compared with other nitrogen sources such as urea and ammonium carbonate [40]. Accordingly, urea was introduced as the best nitrogen source and alanine, followed by glutamic acid, were the best sources of nitrogen among the other amino acids.…”
